摘要
目的:探讨脾虚型FD大鼠胃组织MLCK蛋白及基因表达及脾虚1号方干预。方法:60只10d龄雄性SD大鼠随机分为正常对照组(n=10)、脾虚型FD模型组(n=50)。正常组给予2%蔗糖溶液灌胃,脾虚型FD模型组给予0.1%蔗糖碘乙酰胺蔗糖溶液灌胃,每天0.2mL/只,连续6d。脾虚型FD模型组正常饲料喂养至6周龄后叠加改良小平台站立,连续14d。造模结束后随机分为模型组、多潘立酮组、脾虚1号低、中、高剂量组,每组各10只,连同正常组,分别给予蒸馏水1mL·100g^(-1)·d^(-1)、多潘立酮0.3125mg·100g^(-1)·d^(-1)、脾虚1号方0.1275、0.2550、0.5100g·100g^(-1)·d^(-1),灌胃14d。采用免疫组化、Western blot和RT-PCR方法检测胃体组织MLCK蛋白及mRNA表达量。结果:与正常组比较,模型大鼠胃体组织MLCK蛋白和mRNA表达量显著降低(P<0.01);与模型组比较,脾虚1号方高剂量组MLCK蛋白和mRNA表达量显著升高(P<0.01)。结论:脾虚型FD模型大鼠存在胃体组织MLCK蛋白及基因表达的降低,脾虚1号方健脾促动力的分子机制可能是通过上调MLCK蛋白及基因的表达。
Objective: To investigate the expression of MLCK protein in gastric tissue of FD rats with spleen deficiency type and the intervention by Pixu-1 Formula.Methods: Sixty 10-day-old male SD rat pups were randomly divided into normal control group(n=10) and FD with spleen deficiencymodel group(n=50).The normal control group were gavaged with 0.2mL of 2% sucrose, meanwhile, the FD with spleen deficiency type group were gavaged with 0.2 mL of 0.1% IA in 2% sucrose for 6 days. To postnatal 43 day, the FD with spleen deficiency type group was given modified multiple platform method(MMPM), continued 14 h every day, for 14 days. The FD with spleen deficiency type group were randomly divided into model group(n=10), domperidone group(n=10), low-dose Chineseherbs group(n=10), medium-dose Chineseherbs group(n=10), high-dose Chineseherbs group(n=10). These groups were gavaged with 1mL · 100g^(-1) · d^(-1) of distilled water, 0.3125 mg · 100g^(-1) · d^(-1) of domperidone, 0.1275 g ·100g^(-1) · d^(-1) of Pixu-1 Formula, 0.255 g · 100g^(-1) · d^(-1) of Pixu-1 Formula, 0.51 g · 100g^(-1) · d^(-1) of Pixu-1 Formula respectively, for 14 days. The MLCK protein and mRNA expression in gastric corpus tissues were detected by immunohistochemistry, Westernblotand RT-PCR. Results: Compared with the control group, the amount of MLCK protein and mRNA expression of model group was decreased, and the difference was statistically significant(P<0.01); Compared with the model group, the amount of expression of MLCK mRNA and protein of high-dose Pixu-1 Formula group were increased, and the difference was statistically significant(P<0.01). Conclusion: The expression of MLCK protein and gene were decreased in FD rats model with spleen deficiency type. Furthermore, the molecular mechanism of Pixu-1 Formula invigorating spleen and promoting power might be up-regulating the expression of MLCK protein and gene.
