摘要
建立了高效液相色谱法测定穿苦颗粒中黄芪甲苷含量的方法。采用Waters symmetry C18色谱柱,流动相为水-乙腈(65∶35,V∶V),柱温30℃,流速1.0 m L/min,漂移管温度60℃,雾化器温度36℃,通气气流压力40 psi,黄芪甲苷在浓度1.0-10μg范围内线性良好,相关系数r为0.999 6,平均加样回收率为98.0%,RSD为4.0%。该方法操作简便、稳定性好、结果准确,为穿苦颗粒的质量控制和评价提供了依据。
A method was established for determining of astragaloside in traditional Chinese medicine preparation Chuanku granule by high performance liquid chromatography with evaporative light scattering detector(HPLC-ELSD).Waters symmetry C18 chromatographic column was used with a 1.0 m L/min flow rate in 30 ℃ condition,the mobile phase was water-acetonitrile(65∶35,V∶V).The temperatures of drift tube and atomizer were 60 ℃ and 36 ℃,respectively,stream pressure was 40 psi.The results showed that the calibration curve was linear(r=0.999 6) in the range of 1.0-10.0 μg.The average recovery was 98.0%with RSD of 4.0%.This established method was simple,repeatable and veracious,and it was well suitable for the product quality inspection and control for Chuanku granule.
