摘要
目的:研究以HPLC-ELSD法测定枸杞子中甜菜碱的含量。方法:采用HPLC-ELSD法。Luna5u SCX100A色谱柱(4.6mm×250mm,5μm),流动相甲醇-0.10mol·L-1醋酸铵(15∶85),体积流量1.0mL·min-1,柱温30℃,检测器参数为雾化器温度60℃,蒸发器温度90℃,蒸发器气流1.0L·min-1。结果:甜菜碱线性范围为0.75~3.75μg(r=0.9997),平均回收率为99.86%,RSD为2.05%(n=6)。结论:该方法快速,准确,适用于测定枸杞子中甜菜碱的含量。
Objective:Aim to establish a HPLC-ELSD method for determination of betaine in Lycium barbamm. Method:HPLC-ELSD method was used Luna 5 u SCX 100A(4. 6 mm × 250 mm,5 μm) ,was used with methanol-0. 10 mol·L^-1 ammonium acetate(15∶ 85) ,the flow rate was 1. 0 mL·min^-1,the column temperature was at 30 ℃;Detector parameter were as follow:the nebulizar temperature was at 60 ℃,and the evaporation temperature was at 90 ℃,the flow rate was 1. 0 L·min^-1. Result:The linear range of betaine was in 0. 75 ~ 3. 75 μg(r = 0. 999 7);The average recovery(n = 6) was 99. 86%,with RSD 2. 05% . Conclusion:The method is suitable for the determination the contents of the betaine in L. barbamm.
