摘要
目的:对比研究不同精液优化处理技术的优劣势。方法:选取2018年1月-2019年5月于我院生殖中心就诊,且禁欲3~5d后采集并送检至男科实验室的100份新鲜精液标本作为研究对象,根据不同精子优化处理技术进行分组,SUP组(采用上游法处理)、DGC组(采用密度梯度离心法处理)、联合组(先进行DGC法再进行SUP),比较三种方法处理后精子存活率、活力、形态、DFI及HDS水平。结果:三组处理后精子基本情况比较,联合组精子存活率、活动率均显著高于其余两组, SUP组显著优于DGC组,联合组DFI、HDS值显著低于其余两组,SUP组低于DGC组,差异有统计学意义(P<0.05),三组精子畸形率相当,差异无统计学意义。结论:DGC联合SUP对精子处理具有更好的效果,有利于提高精子存活率,保持DNA完整,减少损伤,在精液优化处理技术中值得推广应用。
Objective:Comparative Study on the Advantages and Disadvantages of Different Semen Processing Technologies.Methods:From January 2018 to May 2019, 100 fresh semen samples were collected from reproductive center of our hospital and sent to andrology laboratory after abstinence for 3~5 days.They were divided into three groups according to different optimized sperm treatment techniques. They were divided into SUP group(using upstream method),DGC group(using density gradient centrifugation)and combined group(using DGC first,and then using SUP).The sperm viability, viability,morphology,DFI and HDS levels were compared between the three groups.Results:The sperm survival rate and motility rate of the combined group were significantly higher than those of the other two groups, the SUP group was significantly better than the DGC group,the DFI and HDS values of the combined group were significantly lower than those of the other two groups,and the SUP group was lower than the DGC group, the difference was statistically significant(P<0.05).The sperm abnormality rate of the three groups was similar, the difference was not significant.Conclusion:Density gradient centrifugation combined with upstream method has a better effect on sperm treatment, which is conducive to improving sperm survival rate, maintaining DNA integrity and reducing damage. It is worth popularizing and applying in semen optimization technology.
