摘要
目的:优化黑根霉胞外多糖摇瓶发酵条件及其体外抗氧化活性的研究。方法:在单因素试验的基础上采用正交试验,考察不同的发酵时间、装液量和转速对黑根霉胞外多糖摇瓶发酵产糖量和真菌生长的影响。通过测定对1,1-二苯基-2-三硝基苯肼(DPPH)自由基、羟基自由基(·OH)的清除能力评价其抗氧化活性。结果:经试验确定黑根霉胞外多糖摇瓶发酵的最佳条件为发酵时间6d、装液量150mL、摇瓶转速160 r/min。在EPS浓度为5 mg/mL时,对DPPH自由基和·OH自由基的清除率分别达到71.65%和76.60%。结论:通过对黑根霉胞外多糖摇瓶发酵条件的优化提高了发酵的生物产量和效率;同时明确了黑根霉胞外多糖有较强的抗氧化能力,试验结果为黑根霉胞外多糖的研究与开发提供了理论依据。
Objective: Optimization of shake flask fermentation conditions and study on the antioxidant activity in vitro of exopolysaccharides from Rhizopus nigricans. Methods: The orthogonal test was carried out based on the single factor test. The effects of different fermentation time, the volume of liquid and shake flask speed on the sugar production and fungal growth of the shake flask fermentation in exopolysaccharides from Rhizopus nigricans were observed. The antioxidant activity of scavenging capacity against DPPH free radical and hydroxyl free radical was determined. Results: The shake flask fermentation conditions of exopolysaccharides from Rhizopus nigricans were optimized as follows: fermentation time 6 d, the volume of liquid 150mL, shake flask speed 160r/min. The clearance rate of DPPH free radical and hydroxyl free radical could reach to 71.65% and 76.60% under the concentration of EPS was 5mg/mL. Conclusion: The fermentation conditions were optimized to improve the exopolysaccharide yield and efficiency of fermentation. And the determination results suggested that the exopolysaccharides from Rhizopus nigricans had strong antioxidant activities. The experimental results provide a theoretical basis for the research and development of the exopolysaccharides from Rhizopus nigricans.
