摘要
目的对1名AB型献血者血液中的抗-Mur抗体系统地进行特异性、反应能力等检测,并利用此抗体对中山市人群Mur抗原发生频率进行调查。方法在本实验室,应用合成的Mur抗原特异性多肽,通过红细胞多肽凝集抑制试验鉴定抗-Mur;英国国际血型参比实验室对该抗体进行了经典血型血清学试验、试管凝集方法和Miltenberger抗原谱细胞鉴定抗-Mur。观察蛋白水解酶、酸碱度对Mur抗原抗体反应的影响,以微量板盐水法检测广东中山地区Mur抗原发生频率。结果确定了该抗体是高质量的Mur血型抗原检测试剂;木瓜酶、菠萝酶能破坏Mur抗原,弱酸性环境能增强Mur抗原抗体反应,中山地区献血者Mur抗原占7.31%(128/1 752)。结论我们发现的这例人源抗-Mur,无ABO天然抗体存在,效价高,特异性强,为理想的Mur血型定型抗体,为今后国内外输血界对Mur血型相关研究提供了基本保证。中山地区Mur抗原发生频率较高,输血时应该予以充分的重视。
Objective To determine the reaction ability and the specificity of an anti-Mur antibody obtained from an type AB blood donor, and to detect the frequency of Mur antigen in population of Zhongshan, China. Methods The anti- body was identified by hemagglutination inhibition test using synthetic Mur antigen specific polypeptide in our laboratory. In parallel, the antibody was identified by classic blood group serological test, tube agglutination method and Miltenberger anti- gen spectrum cell in the Bristol Institute for Transfusion Sciences (BITS) and International Blood Group Reference Laborato- ries (IBGRL). After testing the effects of proteolytic enzymes and pH on the reaction of Mur antigen and antibody, the anti -Mur antibody was used to investigate the Mur antigen frequency in Zbongshan by using hemagglutination assay. Results A rare anti-Mur antibody which specifically recognized Mur (MNSIO) antigen was discovered. Papain and bromelain could in- activate the Mur antigen. Weak acid environment can enhance the reaction of antigen and antibody. The Mur antigen frequen- cy of blood donors in Zhongshan area was 7.31% ( 128/1 752). Conclusion A high titer and specificity of human anti-Mur antibody was found, which contains no ABO antibodies. It is ideal for Mur blood typing, and it will provide a guarantee for further study on Mur blood transfusion. The Mur antigen frequency in Zhongshan is quite high, and more attention should be given to clinical blood transfusion.
