中国部分人群Mur血型抗原分布及分子基础的研究 被引量:18

Distribution and molecular basic study of Mur antigen in different Chinese populations

摘要 目的调查上海地区和云南地区献血者中Mur抗原阳性的分布频率;研究其基因型和分子基础。方法用已知的抗-Mur抗血清在上海和云南少数民族的献血者中用U型微孔板法筛查Mur表型,得出Mur血型抗原表型在这2种人群中的分布频率。对血清学筛出的Mur阳性标本进行Miltenberger血型基因SSP分型,同时进行GYPB假外显子的PCR扩增和直接测序或克隆后测序分析。结果在1 673名上海献血者中筛选出Mur抗原阳性30例,即Mur抗原表型在上海地区中的频率为1.79%;在201名云南献血者中筛选出Mur抗原阳性7例,即Mur抗原表型在云南地区中的频率为3.48%。对实际获得的28份上海献血者和7份云南献血者DNA的分析显示,27份上海献血者阳性标本及6份云南阳性标本均为GYP(B-A-B)Mur,即为GP.Mur型。其余2例Mur抗原阳性献血者DNA测序结果为1种新的GYP(B-A-B)杂交基因型,我们命名为GYP MiⅢShanghai。突变的位点为GYPB上209C>A、220A>G、224A>G、227A>C、内含子3的1T>G,推测产生的蛋白序列与GP.Bun一致。结论本文统计了上海地区和云南地区献血者Mur抗原分布频率;同时发现了1种新的Mur抗原相关GYP(B-A-B)基因型。中国人群中Mur抗原阳性表型对应的基因型至少包括GYP MiⅢ和GYP MiⅢShanghai这2型。 Objective To screen the Mur antigen among the blood donors of Shanghai and Yunnan, and to conduct more molecular studies to confirm its genetic phenotype and molecular character. Methods Mur antigen-positive was detected by micro-plate and known Mur-antibody-positive serum. The results would be calculated into pereentum. SSP methods were performed for Mur genotyping. Based on the Mur antigen's genetic traits, two appropriate primers were designed for PCR ampli- fication. PCR amplification of GYPB was performed. The PCR products were sequenced direcdy or sequenced after cloning. Results There were 30 Mur-antigen-positive samples among the total 1 673 Shanghai's cases at a rate of 1.79%. There were 7 cases among 201 Yunr/an's cases at a rate of 3.48%. A method to amplify the pseudoexon has also been found. 96.4% of Shanghai's Mur-antigen-positive samples and 85.7% ones of Yunnan's were Gp. Mur. However, others were new kinds of genetic phenotype, with nucleotide changes in GYPB 209C 〉 A; 220A 〉 G; 223A 〉 G; 227A 〉 C and intron 3 1T 〉 G. We named it GYP Milll Shanghai. The protein sequence was speculated to be consistent with GP. Bun. Conclusion This study counted the distribution frequency of Mur antigen among the blood donators from Shanghai and Yunnan, and found a new GYP (B-A-B) genetic phenotype related to Mur antigen. There are at least GP. Mur and GP. Bun, two genetic phenotypes in the Mur-antigen-positive Chinese population.
