慢性增强型体外反搏对高胆固醇血症猪血管内皮细胞凋亡的影响 被引量:4

Effect of Long-term EECP on Apoptosis of Vascular Endothelial Cells in Porcine Aorta with Hypercholesterolemia

摘要 【目的】探讨慢性增强型体外反搏对高胆固醇血症猪主动脉血管内皮细胞凋亡的影响。【方法】18头雄性乳猪随机分为正常饲养组(n=6),高脂饲养组(n=6)及高脂饲养+反搏组(n=6)。后2组复制高胆固醇血症猪模型并对高脂饲养+反搏组进行为时36h的慢性增强型体外反搏,收集3组动物主动脉血管内皮细胞,TUNEL法测定其主动脉血管内皮细胞的凋亡指数。【结果】与正常饲养组相比,高脂饲养组和高脂饲养+反搏组血清总胆固醇和低密度脂蛋白明显升高。主动脉血管内皮细胞凋亡指数在正常饲养组为(127±36)‰,在高脂饲养组为(237±23)‰,在高脂饲养+反搏组为(177±12)‰,高脂饲养组和高脂饲养+反搏组较正常饲养组明显升高(P<0.05),高脂饲养+反搏组较高脂饲养组明显降低(P<0.05)。【结论】高胆固醇血症刺激血管内皮细胞凋亡,慢性增强型体外反搏通过减轻血管内皮细胞凋亡拮抗高胆固醇脂血症对血管内皮的损伤作用。 [Objective] To explore the effect of long-term enhanced external counterpulsation (EECP) on apoptosis of vascular endothelial ceils in the aorta of hypercholestemlemic pigs. [ Methods ] Eighteen male infant pigs were randomly divided into 3 groups according to their chow diet: the normal control group (n=6), the hypercholesterolemic control group (n=6) and the hypercholestemlemic + EECP group(n=6). After having performed EECP for 36 hours on the hypercholestemlemic + EECP group(n=6) , we harvested the vascular endothelial ceils from the aorta of three groups of pigs and detected their apoptosis indexes by TUNEL. [Results] The apoptosis indexes in vascular endothehal ceils of both the hypercholesterolemic control group and the hypercholestemlemic + EECP group were significantly higher than that of the normal control group [ (237±23)‰, (177±12)‰ vs. (127 ± 36)‰, respectively, P〈 0.05 ].The apoptosis index of the hypemholestemlemic +EECP group was obviously lower compared with that of the hypercholestemlemic control group (P〈 0.05). [Conclusion] Long-term EECP may protect vascular endothelial cells from the damage resulting from hypercholesterolemia by reducing the apoptosis of vascular endothelial ceils.
