摘要 目的探讨鼻咽癌中DNA聚合酶β(DNApolymeraseβ,polβ)的变异情况。方法采用RT -PCR法(逆转录DNA聚合酶链反应)、PCR SSCP(聚合酶链单链构象多态性分析)、DNA序列分析法对2 6例鼻咽癌组织及癌旁组织标本进行polβ基因突变研究。结果在2 6例癌旁组织标本中polβ无突变;2 6例鼻咽癌组织标本中存在polβ基因变异的有8例,突变率为30 8%。polβ基因变异的鼻咽癌标本测序结果显示:核苷酸序列有多种形式的改变。结论在鼻咽癌中存在多种形式的polβ突变,导致氨基酸序列、二级空间结构的改变。 Objective To study DNA polymeraseβ(polβ) gene mutation in human nasopharyngeal cancer. Methods Twenty-six cases of nasopharyngeal carcinoma tissues and control epithelial tissues were collected to study DNA polβgene mutation with PT-PCR, PCR-SSCP and DNA sequence analyzing technique. Results polβ gene mutation was identified by SSCP in 8 of 26 nasopharyngeal cancer tissues (with mutation rate 30.8%) and there was no mutation in juxtacancerous tissues. Many kinds of mutation were found by DNA sequencing. Conclusion The study proves polβ alternation in human nasopharyngeal carcinoma, this results in sequence alternation in amino acid and spatial structure changing. These mutations may play an important role in carcinogenesis of nasopharygeal cancer.
