兔VX2骨肿瘤生长及转移与辅助性T细胞功能的关系 被引量:1

Relationship between growth and metastasis of VX2 bone tumor and function of T helper cells in rabbits

摘要 目的探讨兔VX2骨肿瘤生长及转移与辅助性T细胞功能的关系。方法将40只日本大耳白兔随机分为4组,A、B、C组分别在植瘤后1、2、3周处死,D组不植瘤,为正常对照组,实验结束前处死。4组在植瘤前及处死前抽取外周血,采用双抗体夹心酶联免疫吸附法检测外周血、肺及骨肿瘤周围软组织中IFN-γ和IL-4浓度,并行组织病理检测。结果在4组植瘤前,外周血IFN-γ、IL-4及IFN-γ/IL-4比值差异无统计学意义(P>0.05),处死前差异有统计学意义(P<0.05);组间比较:B组与A、C、D组差异有统计学意义(P<0.05),C组与A、D组差异有统计学意义(P<0.05);组内比较:B、C组植瘤前与处死前差异有统计学意义(P<0.05)。肺及骨肿瘤周围软组织中IFN-γ、IL-4及IFN-γ/IL-4比值差异有统计学意义(P<0.05),肺组织组间比较:B组与C、D组差异有统计学意义(P<0.05),C组与A、B、D组差异有统计学意义(P<0.05),骨肿瘤周围软组织组间比较:D组与A、B、C组差异有统计学意义(P<0.05),A组与B、C组差异有统计学意义(P<0.05)。结论种植VX2肿瘤后可引起兔外周血、肺及骨肿瘤周围软组织中IFN-γ下降、IL-4上升,Th1和Th2比值失调。 Objective To investigate the relationship between rabbit VX2 bone tumor growth and metastasis and T helper cell function. Methods Forty Japanese white rabbits were randomly divided into four groups. Groups A,B,C were sacrificed at 1,2 and 3 weeks after tumor implantation,respectively,while group D was not implanted and served as the normal control group that was sacrificed toward the end of the experiment. Peripheral blood was collected before tumor implantation and execution in the four groups. The concentrations of IFN-γ and IL-4 in the peripheral blood,lung,and soft tissues around bone tumors were measured by double antibody sandwich enzyme-linked immunosorbent assay,and histopathological examination was performed. Results Peripheral blood IFN-γ,IL-4 and IFN-γ/IL-4 ratios were not significantly different before tumor implantation in the four groups(P>0.05),but there was statistically significant difference before execution(P<0.05). In comparison,the difference between group B and groups A,C,D was statistically significant(P<0.05),so was the difference between group C and groups A,D(P<0.05). There was significant difference between group B and group C before tumor implantation and before death(P<0.05). There was significant difference in IFN-γ,IL-4 and IFN-γ/IL-4 ratios in the lung and soft tissue around bone tumors(P<0.05). In terms of lung tissues the difference between groups B and groups C,D was statistically significant(P<0.05),so was the difference between group C and groups A,B,D(P<0.05). As for the soft tissue surrounding bone tumors,the difference between group D and groups A,B,C was statistically significant(P<0.05),so was the difference between group A and groups B,C(P<0.05). Conclusion Implantation of VX2 tumor can cause the decrease of IFN-γ and the increase of IL-4 in peripheral blood,lungs and soft tissue surrounding bone tumors in rabbits,and the imbalance of Th1/Th2 ratio.
