小鼠肝癌肺转移模型的建立

Establishment of lung metastasis model of liver cancer in mice

摘要 目的探讨通过小鼠尾静脉注射肝癌H22细胞建立小鼠肝癌肺转移模型的可行性。方法2020年9月至2021年1月,笔者根据小鼠尾静脉注射H22细胞数量的不同,将Balb/c小鼠分为对照组(生理盐水200μl)、低浓度组(2×10^(4)个/200μl)和高浓度组(2×10^(5)个/200μl)。每组均于注射瘤细胞后7d、14d、21d处死小鼠,提取小鼠肺脏进行大体及镜下形态观察,并对肺组织进行增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)免疫组织化学染色。结果对小鼠尾静脉注射瘤细胞后7d,结果显示高浓度组与低浓度组小鼠肺组织内成瘤率分别为30%和20%;注射瘤细胞后14d,结果显示高浓度组与低浓度组小鼠肺组织内成瘤率分别为70%和40%;注射瘤细胞后21d,结果显示高浓度组小鼠肺组织内成瘤率为90%,明显高于7d成瘤率,差异有统计学意义(P=0.006,P<0.01),而低浓度组小鼠肺组织内成瘤率仅为60%。瘤结节大小不一,散在分布于肺组织内。21d时瘤细胞PCNA增殖指数在40%~50%。对照组小鼠肺组织内未见肿瘤形成。结论经小鼠尾静脉注射H22肝癌细胞制备肝癌肺转移模型,有成瘤率高、周期短、操作简单、成本低等特点,且形态学特点与人体转移瘤相近,可作为肿瘤研究的模型之一。 Objective To explore the feasibility of establishing lung metastasis model of liver cancer by the tail vein injection of liver cancer cells H22 in mice.Methods From September 2020 to January 2021,the Balb/c mice were divided into control group(200μl normal saline),low concentration group(2×10^(4)pieces/200μl),high concentration group(2×10^(5)pieces/200μl)according to the number of cells by tail vein injection.The mice were sacrificed on day 7,day 14 and day 21 after injection of tumor cells,and the lung was taken for gross and microscopic examination.Immunohistochemical staining for proliferating cell nuclear antigen(PCNA)was performed in the lung tissue.Results Seven days after injection of tumor cells,the tumor formation rates in lung tissues in the high concentration group and the low concentration group were 30%and 20%,respectively.Fourteen days after injection,the tumor formation rates in the high concentration group and the low concentration group were 70%and 40%,respectively.Twenty-one days after tail vein injection,the tumor formation rates of the high concentration group was 90%,which was significantly higher than 7d(P=0.006,P<0.01).The tumor formation rate in the low concentration group was only 60%.Tumor nodules varied in size and were scattered in the lung tissues.On day 21,PCNA proliferative index of tumor cells in the lung tissue was between 40%-50%.There was no tumor formation in the lung tissue of the control group.Conclusion Pulmonary metastasis of liver cancer model might be achieved via tail vein injection of liver cancer cells,which possesses high tumor formation rate,featuring short duration,simple manipulation and low cost.Moreover,the morphologic features are similar to those of human metastatic carcinoma.It could be used as a model for the investigation of tumor.
