四君子汤对三阴性乳腺癌细胞及小鼠模型增殖、转移的抑制作用 被引量:3

Inhibition of Sijunzi Decoction on the proliferation and metastasis of triple negative breast cancer cells and mice models

摘要 目的:探索四君子汤对三阴性乳腺癌(TNBC)细胞及小鼠模型增殖、转移的抑制作用。方法:采用CCK-8法、细胞划痕实验、Transwell实验和Western Blot技术分别检测不同浓度的四君子汤对小鼠TNBC 4T1细胞的细胞活力、细胞伤口愈合能力、细胞迁移能力及转移相关蛋白表达的影响。18只小鼠随机分为空白组、模型组和四君子汤治疗组。模型组、四君子汤治疗组接种4T1细胞建立TNBC移植瘤小鼠模型。四君子汤治疗组小鼠按4.85 g·kg^(-1)·d^(-1)灌胃四君子汤剂;空白组和模型组小鼠灌胃等量0.9%氯化钠溶液,并实时监测小鼠的肿瘤大小。采用HE染色检测小鼠癌细胞肺转移情况。结果:与对照(0 mg/mL)组比较,四君子汤具有抑制4T1细胞增殖的作用,且呈时间和浓度依赖性。与对照(0 mg/mL)组比较,5、10 mg/mL四君子汤能够显著抑制4T1细胞的伤口愈合及迁移能力(P<0.05,P<0.01);10 mg/mL四君子汤可上调4T1细胞中E-cadherin蛋白表达水平(P<0.05),2.5、5 mg/mL四君子汤可下调β-catenin蛋白表达水平(P<0.01,P<0.05)。与模型组比较,四君子汤治疗组小鼠17 d后移植瘤体积显著减小(P<0.05),肺转移情况明显改善。结论:四君子汤能够抑制TNBC的增殖和转移。 Objective:To explore the inhibitory effect of Sijunzi Decoction on the proliferation and metastasis of triple negative breast cancer(TNBC)cells and mice models.Methods:CCK-8 assay,cell scratch assay,Transwell assay and Western Blot were used to detect the effects of Sijunzi Decoction of different mass concentrations on cell viability,wound healing ability,cell migration ability and protein expression related to metastasis of mouse three-negative breast cancer 4T1 cells.A total of 18 mice were randomly divided into control group,model group and Sijunzi Decoction group.Model group and Sijunzi Decoction group were inoculated with 4T1 cells to establis TNBC transplanted tumor model.Sijunzi Decoction group were given Sijunzi Decoction by gavage according to 4.85 g•kg^(-1)•d^(-1).Control group and model group were given the same amount of saline.Their tumor size were monitored in real time.HE staining was used to detect lung metastasis of cancer cells in mice.Results:Compared with the control(0 mg/mL)group,Sijunzi Decoction inhibited the proliferation of 4T1 cells,which was time-dependent and concentration-dependent.Compared with the control(0 mg/mL)group,5,10 mg/mL Sijunzi Decoction could inhibit the wound healing and migration of 4T1 cells(P<0.05,P<0.01),and 10 mg/mL Sijunzi Decoction could up-regulate the expression of E-cadherin protein(P<0.05),and 2.5,5 mg/mL Sijunzi Decoction could down-regulateβ-catenin protein in 4T1 cells(P<0.01,P<0.05).Compared with the model group,after the treatment of 17 days,the tumor volume of the transplanted mice in Sijunzi Decoction group was significantly reduced(P<0.05),and the lung metastasis was improved.Conclusion:Sijunzi Decoction may inhibit the proliferation and metastasis of TNBC.
