四君子汤对脾虚小鼠感染念珠菌后血清中IFN-γ含量及腹腔巨噬细胞吞噬功能的影响 被引量:8

摘要 目的研究小鼠在脾虚时经口感染白念珠菌后经四君子汤灌胃治疗,检测小鼠血清中IFN-γ含量的变化及腹腔巨噬细胞的吞噬功能,探讨脾虚小鼠在感染白色念珠菌经中药治疗后的免疫机制。方法选取SPF级昆明种小白鼠,随机分组,经口感染白念珠菌,中药四君子汤灌胃治疗,检测腹腔巨噬细胞吞噬白色念珠菌的吞噬百分率,用酶联免疫吸附(ELISA)法测定小鼠血清中γ干扰素(IFN-γ)的含量;结果与正常对照组比较,脾虚模型组、脾虚+白色念珠菌组、脾虚+白色念珠菌+氟康唑组、脾虚+白色念珠菌+四君子汤组小鼠腹腔巨噬细胞吞噬白色念珠菌的吞噬率在给药的第14天显著降低(P<0.01)。与脾虚模型组比较,脾虚+白色念珠菌组、脾虚+白色念珠菌+四君子汤组小鼠腹腔巨噬细胞吞噬白色念珠菌的吞噬率在给药的第14天均降低(P<0.05),与脾虚+白色念珠菌组比较,脾虚+白色念珠菌+氟康唑组、脾虚+白色念珠菌+四君子汤组小鼠腹腔巨噬细胞吞噬白色念珠菌的吞噬率在给药的第14天显著升高(P<0.01)。小鼠血清中IFN-γ的含量,与正常对照组比较,脾虚+白色念珠菌+氟康唑组、脾虚+白色念珠菌+四君子汤组小鼠血清中IFN-γ的含量在给药的第14天含量显著升高(P<0.01)。与脾虚模型组比较,脾虚+白色念珠菌+氟康唑组、脾虚+白色念珠菌+四君子汤组小鼠血清中IFN-γ的含量在给药的第14天含量明显升高(P<0.05或P<0.01)。与脾虚+白色念珠菌组比较,脾虚+白色念珠菌+四君子汤组小鼠血清中IFN-γ的含量在灌胃四君子汤的第14天含量明显升高(P<0.05)。结论当机体在脾虚状态下,经消化道感染白念珠菌后会导致免疫力下降,而四君子汤通过益气健脾,增强机体免疫力,起到治疗作用。 Objective To study the changes of IFN-γin serum and the phagocytic function of peritoneal macrophages in spleen deficiency mice infected with Candida albicans by oral administration of Sijunzi Decoction and explore the immune mechanism of mice infected with Candida albicans after treatment with traditional Chinese medicine.Methods SPF Kunming mice were randomly divided into groups.Candida albicans was infected orally.Sijunzi Decoction was given orally.The phagocytosis percentage of peritoneal macrophages phagocytizing Candida albicans was detected.The content of interferon-γ(IFN-γ)in serum of mice was determined by enzyme-linked immunosorbent assay(ELISA).Results Compared with that of the normal control group,the phagocytosis rate of peritoneal macrophages of mice in spleen deficiency model group,spleen deficiency+Candida albicans group,spleen deficiency+Candida albicans+fluconazole group and spleen deficiency+Candida albicans+Sijunzi Decoction group decreased significantly on the 14 th day of administration(P<0.01).Compared with that of the model group of spleen deficiency,the phagocytosis rate of peritoneal macrophages of mice in the spleen deficiency+Candida albicans group and the spleen deficiency+Candida albicans+Sijunzi Decoction group decreased on the 14 th day of administration(P<0.05).Compared with that of the spleen deficiency+Candida albicans+fluconazole group,spleen deficiency+Candida albicans+Sijunzi Decoction group,the peritoneal macrophages of mice in the spleen deficiency+Candida albicans+Sijunzi Decoction group phagocytized Candida albicans.The phagocytosis rate of bacteria increased significantly on the 14 th day of administration(P<0.01).Compared with that of the normal control group,the serum levels of IFN-γof mice in the spleen deficiency+Candida albicans+fluconazole group and the spleen deficiency+Candida albicans+Sijunzi Decoction group increased significantly on the 14 th day of administration(P<0.01).Compared with that of the model group of spleen deficiency,the levels of IFN-γin serum of mice in spleen deficiency+Candida albicans+fluconazole group and spleen deficiency+Candida albicans+Sijunzi Decoction group increased significantly on the 14 th day of administration(P<0.05 or P<0.01).Compared with that of the spleen deficiency+Candida albicans group,the serum IFN-γcontent of mice in the spleen deficiency+Candida albicans+Sijunzi Decoction group increased significantly on the 14 th day of intragastric administration of Sijunzi Decoction(P<0.05).Conclusion When the body is in the state of spleen deficiency,Candida albicans infection through the digestive tract will lead to a decline in immunity,and Sijunzi Decoction,through invigorating Qi and strengthening the spleen and enhancing the body’s immunity,plays a therapeutic role.
