白头翁皂苷B4对临床型乳房炎奶牛乳汁体细胞、酶、炎性和抗氧化因子影响 被引量:7

摘要 【目的】乳房炎是危害奶牛健康最严重的疾病,常造成奶牛乳汁体细胞数升高和细胞因子水平改变。白头翁皂苷B4是中药白头翁的主要成分,具有抑菌、抗炎、抗氧化和增强免疫力等功能。对临床型乳房炎奶牛注射白头翁皂苷B4注射液,为探究白头翁皂苷B4对临床型乳房炎奶牛乳汁SCC、酶、炎性和抗氧化因子的影响,为临床应用提供依据。【方法】选取规模化奶牛场2-3胎次泌乳高峰期健康中国荷斯坦奶牛(对照组,不注射药物)和临床型乳房炎奶牛(试验组)各12头。乳房炎发病当天记为第0天,按0.05 mL/kg连续颈部肌肉注射白头翁皂苷B4注射液6 d,1次/d,并观察临床疗效至第12天。分别使用体细胞检测仪和ELISA法检测各组奶牛第0、1、3、5、7、9、11天乳汁SCC和乳清酶LDH、NAG、ALT、AST、ALP、ACP,炎性因子IL-1β、IL-6、IL-8、TNF-α,抗氧化因子T-AOC、GSH-Px、SOD、CAT、MDA含量,并应用SPSS 24.0软件进行统计学分析。【结果】临床型乳房炎奶牛乳汁SCC,乳清酶LDH、NAG、ALT、AST、ALP、ACP,炎性因子IL-1β、IL-6、IL-8、TNF-α、脂质过氧化物MDA含量均显著高于对照组(P<0.05),并随治疗时间推移显著下降(P<0.05);而抗氧化因子T-AOC、GSH-Px、SOD、CAT含量显著低于对照组(P<0.05),并随治疗时间推移显著上升(P<0.05);第11天时除ALP显著高于对照组外(P<0.05),其余指标均与对照组差异不显著(P<0.05)。【结论】白头翁皂苷B4可显著降低临床型乳房炎奶牛炎症反应和乳汁SCC,促进乳酶恢复正常水平,增强乳腺抗氧化能力,是治疗奶牛临床型乳房炎的理想药物。 [Objective]Mastitis is the most serious disease that endangers the health of dairy cows and leads to the increase of somatic cell counts and variations of cytokine level.Pulsatilla Saponin B4 is the main component of Pulsatilla,which has antibacterial,anti-inflammatory,anti-oxidation and immunity-enhancing functions.To study the effects of Pulsatilla saponin B4 on somatic cell counts,enzyme,inflammatory and antioxidant factors in milk of dairy cows with clinical mastitis and provide the theoretic foundation for clinical application,Pulsatilla Saponin B4 was administrated to treat dairy cows with clinical mastitis.[Method]Twelve healthy dairy cows(control group,without administration)and 12 clinical mastitis cows(test group)were selected in the experiment.The onset day of mastitis was recorded as the 0th day,the cows with clinical mastitis were injected with 0.05 mL/kg Pulsatilla saponins B4 once a day for 6 days through cervical intramuscular injection,and the clinical efficacy was observed until the 12th day.The contents of SCC,enzyme LDH,NAG,ALT,AST,ALP,ACP,inflammatory factors IL-1β,IL-6,IL-8,TNF-α,antioxidant indexes T-AOC,GSH-Px,SOD,CAT and MDA in milk of dairy cows on day 0,1,3,5,7,9,11 were detected by somatic cell analyzer and ELISA.SPSS 24.0 software was used for statistical analysis.[Result]The results showed that the contents of SCC,enzyme LDH,NAG,ALT,AST,ALP,ACP,inflammatory factors IL-1β,IL-6,IL-8,TNF-αand MDA in the test group were significantly higher than those in the control group(P<0.05),and decreased significantly within treatment(P<0.05).The contents of antioxidant factors T-AOC,GSH-Px,SOD and CAT in the test group were significantly lower than those in the control group(P<0.05),and increased significantly within treatment(P<0.05).On the 11th day,all the factors in the test group showed no significant differences comparing with the control group(P>0.05)except ALP(significantly higher than in the control group).[Conclusion]These results suggested that the Pulsatilla saponins B4 can significantly reduce the inflammatory reaction and milk SCC,promote the recovery of milk enzymes,enhance the antioxidant capacity of mammary gland,hence an ideal medicine for clinical cows’mastitis in veterinary clinic.
