不同灭菌方法对桑叶超微粉中芦丁含量的影响 被引量:4

摘要 目的探讨不同灭菌方法对桑叶超微粉中芦丁含量的影响。方法对同批桑叶超微粉分别采用干热灭菌、湿热灭菌、^(60)Co辐照灭菌、微波灭菌、臭氧灭菌和环氧乙烷气体灭菌方法灭菌;采用高效液相色谱法测定灭菌前后的样品中芦丁含量,色谱柱为Thermo Hypersil ODS C_(18)柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.5%磷酸溶液(梯度洗脱),流速为1.0 m L/min,检测波长为358 nm,柱温为25℃,进样量为10μL。结果6种灭菌方法均可达到灭菌效果,但不同灭菌方法对桑叶超微粉中芦丁含量的影响不同。湿热灭菌的桑叶超微粉色泽差,呈深黄绿色且结块;^(60)Co辐照灭菌和微波灭菌的桑叶超微粉中芦丁含量分别为0.1554%和0.1528%,RSD分别为1.86%和1.62%。结论不同灭菌方法对桑叶超微粉中的芦丁含量均有影响,^(60)Co辐照灭菌和微波灭菌方法对芦丁含量影响较小,结合实际生产,宜采用微波灭菌方法对桑叶超微粉进行灭菌。 Objective To investigate the effects of different sterilization methods on the content of rutin from ultra-fine powder of Folium Mori.Methods The same batch of ultra-fine powder of Folium Mori was sterilized by dry heat sterilization,wet heat sterilization,^(60)Co radiation sterilization,microwave sterilization,ozone sterilization and ethylene oxide gas sterilization respectively,and the content of rutin in the samples before and after sterilization was determined by the high-performance liquid chromatography(HPLC)method.The chromatographic column was Thermo Hypersil ODS C18 column(250 mm×4.6 mm,5μm),the mobile phase was methanol-0.5%phosphoric acid with gradient elution,the flow rate was 1.0 m L/min,the detection wavelength was 358 nm,the column temperature was 25℃,and the injection volume was 10μL.Results The six sterilization methods had sterilization effects,but different sterilization methods had different effects on the content of rutin from ultra-fine powder of Folium Mori.The ultra-fine powder of Folium Mori sterilized by wet heat sterilization had poor color,dark yellow green,and caking.The contents of rutin from ultra-fine powder of Folium Mori sterilized by^(60)Co radiation sterilization and microwave sterilization were 0.1554%and 0.1528%with RSDs of 1.86%and 1.62%,respectively.Conclusion Different sterilization methods have effects on the content of rutin from ultra-fine powder of Folium Mori,among which^(60)Co radiation sterilization and microwave sterilization have less effect on the content of rutin.Combined with the actual production,the microwave sterilization method should be selected to sterilize ultra-fine powder of Folium Mori.
