青蒿琥酯对结核分枝杆菌诱导的TNF-α、IL-6表达的影响 被引量:4

The Effect of Artesunate on the Expression of TNF-α and IL-6 Induced by Mycobacterium Tuberculosis

摘要 目的探讨青蒿琥酯(artesunate, ASN)对结核分枝杆菌(mycobacterium tuberculosis, Mtb)诱导THP-1细胞分泌TNF-α和IL-6的影响及机制研究。方法体外培养THP-1细胞。MTT法检测ASN对细胞的活性影响;RT-qPCR检测细胞因子TNF-α、IL-6和TLR2mRNA表达;ELISA检测TNF-α、IL-6分泌,Western blot检测TLR2受体表达。结果 MTT法检测ASN浓度在0~20μg/mL范围内培养48h均对细胞无毒性;与对照组相比,Mtb显著增加TNF-α、IL-6mRNA及蛋白表达,而20μg/mL ASN能显著抑制TNF-α、IL-6 mRNA及蛋白表达(P<0.05)。此外,20μg/mLASN能显著抑制TLR2mRNA及蛋白表达。结论 ASN可能通过抑制TLR2受体表达缓解Mtb诱导的炎症反应。 Objective To investigate the effect of artesunate(ASN)on the secretion of cytokine TNF-α and IL-6 in THP-1 cells induced by Mtb and to investigate its potential underline mechanism. Methods THP-1 cells were cultured in vitro.MTT assay was used to detect the effect of ASN on the activity of cells.RT-qPCR was used to detect the mRNA expressions of TNF-α,IL-6 and TLR2.ELISA was used to detect the secretion of TNF-α and IL-6 protein.Western blot was used to detect the protein expression of TLR2. Results There was no cytotoxicity to cell dealth with the concentrations of 0-20ug/ml of ASN during 48hculture.Compared with the control group,Mtb significantly increased mRNA and protein expressions of TNF-α and IL-6.ASN could inhibit the secretion of TNF-α and IL-6 at mRNA and protein levels( P <0.05).In addition,ASN could down-regulate the expressions of TLR2 mRNA and protein. Conclusion ASN may inhibit the inflammatory responses induced by Mtb through the down-regulation of TLR2 expression.
