青蒿琥酯及二氢青蒿素逆转耐药结核分枝杆菌的初步研究 被引量:14

Preliminary Study of Artesunate and Dihydroartemisinin Reversing the Susceptibility of RFP or INH Resistant Mycobacterium tuberculosis

摘要 【目的】了解青蒿素类中药单体(二氢青蒿素、青蒿琥酯)单独或联合利福平(RFP)及异烟肼(INH)对耐药结核分枝杆菌的作用,初步研究此类单体是否具有逆转细菌耐药性的作用。【方法】刃天青显色法判断各中药单体是否有抑菌或杀菌作用;MB/BACT系统(生物梅里埃)检测RFP或INH联合中药单体是否有逆转细菌耐药的作用。【结果】单独使用时各单体均不能杀灭结核分枝杆菌;RFP联合青蒿琥酯(200μg/mL)或二氢青蒿素(200μg/mL)可使11株耐RFP菌株中的9或6株变为敏感,敏感率分别为82%或55%;INH联合上述中药单体则亦可使11株耐INH菌株中的10或7株变为敏感,敏感率分别为90%或64%。【结论】青蒿琥酯及二氢青蒿素有较强的协同相应抗结核药或逆转结核菌的耐药性的作用,但青蒿琥酯敏感率较二氢青蒿素更高。 [ Objective ] To investigate the effect of derivatives from Chinese herbs including artesunate and dihydroartemisinin combined with or without antibiotics such as RFP or INH against RFP-resistant Mycobacterium tuberculosis (RFP-R) or INH- resistant Mycobacterium tuberculosis (INH-R). [Methods] Resazurin colorimetric method and biomeriux MB/BACT MP system were used. [ Results] Both artesunate and dihydroartemisinin can not prevent TB growth alone in MiddteBrook 7H9 media; Artesunate (200 μg/mL) or dihydroartemisinin (200 μg/mL) combined with RFP (2 μg/mL) may sensitive with RFP-resistant isolates (9/11, 6/11 isolates respectively), sensitivity was 82% or 55%, respectively. Artesunate or dihydroartemisinin combined with INH (1 μg/mL) may sensitive INH-R isolates (10/11, 7/11 isolates respectively), sensitivity was 90% or 64%, respectively. [Conclusion] Dihydroartemisinin and artesunate have good effects on reversing RFP or INH resistant- Mycobacterium tuberculosis, and the sensitivity of artesunate is higher than dihydroartemisinin.
