摘要
目的通过分析氯化两面针碱(NC)对体外培养肝癌细胞蛋白表达谱的影响,探讨其对肝癌细胞抑制的作用机制。方法应用蛋白芯片飞行时间质谱技术测定NC作用前后肝癌细胞蛋白的表达谱,在蛋白质专家网站(ExPASy)上进行比对,采用单因素方差分析比较不同组中相同质荷比的蛋白质含量,对处理前后的质谱变化进行配对t检验分析。结果采用NC处理人肝癌细胞SMMC-7721后,应用蛋白芯片飞行时间质谱技术共筛选出有统计学意义的差异表达蛋白质峰14个,这些差异蛋白主要跟细胞凋亡、细胞周期、信号转导、转录、DNA复制及修复、代谢、免疫等通路有关。结论氯化两面针碱对肝癌细胞的抑制作用,可能是通过免疫、代谢等多条通路综合作用的结果。
Objective To analyze the effects of nitidine chloride(NC) on the protein expression profiles of hepatoma in vitro and explore its mechanism of inhibition. Methods The protein expression profiles of hepatoma cells before and after NC treatment were detected by surface-enhanced laser desorption/ionization time of flight mass spectrometry and compared on the ExPASy website. One-way ANOVA was used to compare the protein contents of different groups in the same mass-to-charge ratio.Paired t-test analysis was performed on changes of proteomic spectra. Results After treatment of NC,a total of 14 differentially expressed protein peaks in human hepatoma cells SMMC-7721 were screened by surface-enhanced laser desorption/ionization time of flight mass spectrometry. These 14 proteins were mainly related to apoptosis,cell cycle,signal transduction,transcription,DNA replication and repair,metabolism,and immunity. Conclusion The inhibitory effects of nitidine chloride on hepatoma cells may be through the crosstalk between multiple pathways such as immunity and metabolism.
