子痫前期大鼠模型的建立及相关指标监测 被引量:5

Establishment of rat model of preeclampsia and monitoring of related indexes

摘要 目的:采用减少子宫灌注压(RUPP)的方式建立子痫前期(PE)大鼠模型,检测血压、蛋白尿等生理指标和胎盘相关基因表达。方法:将妊娠大鼠随机分为假手术组(Sham组)和手术组(RUPP组)。合笼见栓第14天,RUPP组于腹主动脉分支子宫动脉处置入银夹,假手术组(Sham组)仅分离血管,不放置银夹。检测手术前后孕鼠血压、尿蛋白和胎盘血流变化,分析胎盘、肾脏等组织的病理学改变和PE相关因子的表达改变。结果:RUPP组术前平均动脉压、尿微量白蛋白/肌酐比值相对平稳,术后开始升高,并显著高于Sham组,于产前达到峰值,产后恢复至术前水平,差异均有统计学意义(P均<0.05)。Sham组术前、术后平均动脉压、尿微量白蛋白/肌酐比值相对平稳,无明显变化。脐动脉血流检测结果示,RUPP组较Sham组的S/D值明显增高,差异有统计学意义(P均<0.05)。胎盘组织HE染色结果示,RUPP组胎盘血管发生不足,迷路层血管孔径较小。肾脏PAS染色结果显示,RUPP组肾小球内皮细胞增生显著异常。RUPP组胎盘组织相关因子表达异常,s Flt-1、HIF-1、PLGF表达均增高,VEGF表达降低,差异均有统计学意义(均P<0.05)。结论:RUPP法诱导的PE大鼠模型是研究PE发病机制及临床干预方法的良好模型。 Objective: To establish a preeclampsia rat model by reduced uterine perfusion pressure( RUPP) method.To detect the expression of placenta related genes and physiological indexes,such as blood pressure,proteinuria etc. Methods: Pregnant rats were randomly divided into two groups,Sham and RUPP. At 14 days of postcoitum( dpc),pregnant rats in RUPP group were tied with silver clip around the uterine artery,the branches of the abdominal aorta.In the Sham operation group,only blood vessels were separated,but no silver clips were placed.The changes of blood pressure,urine protein and placental blood flow in the pregnant rats were analyzed before and after the operation. The histological changes of placenta and kidney,and the expression of preeclampsia-related genes were analyzed.Results: Preoperative mean arterial blood pressure,urinary albumin/creatinine ratio of RUPP group was relatively stable,postoperative mean arterial blood pressure,urinary albumin/creatinine ratio increased,and weresignificantly higher than that of sham,the values reached the peak in the prenatal,postpartum restore to the preoperative level,the differences were statistically significant( both P0.05).The mean arterial pressure and urinary albumin/creatinine ratio were relatively stable in group Sham before and after operation,and there was no significant change. By testing the flow velocity of umbilical artery,the S/D value of RUPP group were increased,the difference was statistically significant( both P 0. 05). Placental tissue HE staining showed that the placental vessels in RUPP group were insufficient,and the diameter of the vessels in the labyrinthine layer was smaller.Renal PAS staining showed that glomerular endothelial cell proliferation was abnormal in group RUPP.Gene expression analysis showed that the expression of placental tissue related factors was abnormal in group RUPP,and the expressions of sF lt-1,HIF-1 and PLGF were increased,and the expression of VEGF was decreased,the difference was statistically significant( both P0.05).Conclusion: The RUPP induced preeclampsia model is a good model for studying the pathogenesis and clinical intervention of preeclampsia.
