康复新液治疗实验性胃溃疡的作用机制研究 被引量:23

摘要 目的:探讨康复新液对实验性胃溃疡大鼠的治疗作用及机制。方法:分别建立束水应激法致急性胃溃疡大鼠模型和乙酸灼烧法致慢性胃溃疡大鼠模型;将健康SD大鼠按体质量分层随机分为空白组、模型组、阳性对照组和康复新液高、中、低剂量组,空白组和模型组灌胃等体积纯水,康复新液各剂量组分别按20、10、5mg·kg^(-1)·d^(-1)灌胃康复新液原药;束水应激法试验中阳性对照组灌胃硫糖铝0.5 g·kg^(-1)·d^(-1),乙酸烧灼法试验中阳性对照组灌胃奥美拉唑2.09 mg·kg^(-1)·d^(-1);分别给药7 d和14 d。给药结束后,对束水应激法建模大鼠测定各组大鼠胃溃疡指数和胃黏膜形态学变化,Elisa法测定血清中5-羟色胺(5-HT)、肿瘤坏死因子ɑ(TNF-ɑ)、胃动素(MTL)、胃泌素(GAS)水平;对乙酸烧灼法建模大鼠检测胃组织形态学,试剂盒测定血清中总超氧化物岐化酶(T-SOD)、一氧化氮(NO)和丙二醛(MDA)水平,胃组织中血管内皮细胞生长因子(VEGF)和前列腺素E_2(PGE_2)的表达。结果:两种胃溃疡模型大鼠的胃黏膜均可见大面积溃疡点,病理检测显示胃黏膜缺损的溃疡样变化,提示模型成功。在急性胃溃疡试验中,与模型对照组比较,康复新液各剂量组大鼠血清中5-HT、TNF-α含量显著降低(P<0.05),GAS、MTL含量具有一定的升高趋势。在慢性胃溃疡试验中,与模型对照组比较,康复新液各剂量组大鼠血清中MDA含量显著降低(P<0.05),NO、T-SOD含量呈升高趋势,胃组织中VEGF、PGE_2含量均升高。结论:康复新液对急性和慢性胃溃疡大鼠均可改善胃黏膜损伤,作用机制与调节胃肠激素、改善胃黏膜血流量和抗氧化有关。 Objective : To observe the therapeutic effect of Kangfuxin Solution on experimental gastric ulcer and further explore its possible mechanism. Method: The acute gastric ulcer model was established by the method of restraint water - immersion stress and chronic gastric ulcer model was established by acetic acid. Health SD rats were randomly divided into control group, model group, positive group ( Sueralfate Chewable Tablets used in restraint water - immersion stress model experiment and Omeprazole used in acetic acid model experiment) and Kangfuxin Solution high( 20 mL ·kg^-1·d^-1) , mediate(10 mL·kg^-1·d^-1) and low(5 mL ·kg^-1·d^-1) doses group by weight. Acute gastric ulcer experiment was administered for 7 days and chronic gastric ulcer experiment was administered for 14 days. Ulcer index was measured after the end of the experiment on restraint water - immersion stress, observing gastric histology by HE staining and determining 5 - HT, TNF - α, MTL and GAS by ELISA assay. Histology of gastric tissue was observed by HE staining on acetic acid model experiment and T - SOD, NO and MDA levels in serum and VEGF as well as PGE2 contents in gastric tissue were determined by Elisa analysis. Result : The results revealed there was a large scale of ulcerous spot and imputed ulcerlike changes by pathological assay in gastric mucosa. In acute gastric ulcer experiment, compared with model group, Kangfuxin Solution remarkably reduced 5 - HT and TNF - α content and promoted GAS as well as MTL expresssion in serum. In chronic gastric ulcer experiment, compared with model group, Kangfuxin Solution significantly decreased the MDA level, and increased the NO and T - SOD levels in serum and elevated VEGF as well as PGE2 levels in gastric tissue. Conclusion: Kangfuxin Solution had significant therapeutic effect on gastric ulcer, involved in regulating gastrointestinal hormone, repairing damaged gastric mucosa, ameliorating blood velocity in mucosa and antioxidative action.
