摘要
目的:研究大鼠实验性胃溃疡自愈期间胃黏膜5-羟色胺(5-HT)和神经肽Y(NPY)的表达变化及其在大鼠实验性胃溃疡自愈中的作用。方法:胃前壁黏膜下注射冰乙酸制备大鼠乙酸性胃体溃疡模型,应用免疫组织化学S-P法检测42只胃溃疡组、21只盐水组及6只正常组大鼠胃黏膜5-HT和NPY的表达,以图像分析系统进行积分光密度值(IOD)测量。结果:正常组大鼠胃黏膜5-HT和NPY蛋白呈弱阳性表达。5-HT阳性细胞主要表达于内分泌细胞,散在分布于胃底腺;NPY主要表达于胃底腺壁细胞、内分泌细胞胞浆内,以靠近黏膜肌层信号较强。溃疡术后1d 5-HT阳性细胞和NPY阳性细胞即显著增多(与正常组相比,P<0.01),至溃疡术后4d 5-HT和NPY积分光密度值均达到高峰,之后虽呈下降趋势,但5-HT阳性细胞积分光密度值仍维持于较高水平(与对照组相比,P<0.01);NPY阳性细胞于溃疡术后23d积分光密度值显著降低,但高于对照组(P<0.01)。结论:大鼠胃溃疡自愈时期5-HT和NPY参与了胃黏膜修复过程。
Objective:To investigate the expression and effect of 5-HT and NPY in gastric mucosa during healing of experimental gastric ulcer for rats. Methods: Ulcers were duplicated by acetic acid injection to submucosa of gastric anterior wall. The expressions of 5-HT and NPY in 42 rats with gastric ulcer(GU),21 rats with normal salinel(NS) and 6 normal rats were detected by immunohistochemical(IHC)SP method. And with image analysis system software,integral optical density value(IOD) was analyzed. Results: The results of IHC showed that expressions of 5-HT and NPY peptide in N group were weakly positive and 5-HT was expressed in cytoplasm of endocrine cells which distributed in fundic glands dispersely. NPY was mainly expressed in cytoplasm of parietal cells and endocrine cells in fundic glands and showed more stronger expressions near muscularis mucosa. On the first day after acetic acid injection, 5-HT and NPY positive cells significantly increased(vs normal group,P〈0.01) ,and reached the peak on the 4th day. Then 5-HT and NPY positive cells showed decreasing, but expression of 5-HT maintained a higher level(vs control group,P〈0.01). The expression of NPY showed significant decreasing on the 23rd day after acetic acid injection,but was more than that in the control group(vs control group, P〈0.01). Conclusion:5-HT and NPY take part in the regulation in renovation to gastric mucosa during healing of experimental gastric ulcer for rats.
