摘要
目的评价枸杞多糖(LBP)及LBP配伍茶多酚(TP)缓解小鼠体力疲劳作用的效果,并探讨其作用机制。方法 SPF级雄性昆明小鼠336只,共分为4批,每批随机分成7组:LBP低、中、高剂量(LBP-L、LBP-M、LBP-H)组,LBP配伍TP(以下简称配伍组)低、中、高剂量(C-L、C-M、C-H)组,设生理盐水(NS)为对照,每组12只。小鼠在每日饲喂基础饲料的基础上,以每10 g体质量0.2 m L溶液灌胃,15 d后进行负重游泳实验,并检测肝糖原、血清尿素氮和血乳酸等生化指标。结果同NS组相比,LBP-M、LBP-H组小鼠负重游泳时间显著延长(P<0.05),LBP-L、LBP-M、LBP-H组肝糖原的含量显著增加(P<0.05),血清尿素氮和血乳酸水平显著降低(P<0.05);C-L、C-M、C-H组小鼠负重游泳时间显著延长(P<0.05),肝糖原的含量显著提高(P<0.05),血清尿素氮和血乳酸水平显著降低(P<0.05)。同LBP组相比,配伍组负重游泳时间延长,肝糖原含量增加,血尿素氮和血乳酸水平降低;其中C-L组肝糖原水平显著高于LBP-L组(P=0.027),C-M组血乳酸水平显著低于LBP-M组(P=0.006)。结论 LBP及LBP配伍TP对小鼠体质量无影响,但具有缓解小鼠体力疲劳的作用;其作用机制可能与其增加了肝糖原储备量、减少蛋白质的分解代谢,进而减少血清尿素氮水平及增加了磷酸果糖激酶的活性,使血乳酸水平下降有关。
Objective To evaluate the effects of Lycium barbarum polysaccharides( LBP) and LBP combined with tea polyphenols( TP) on relieving the physical fatigue in mice and discuss relevant m echanisms. Methods Three hundreds and thirty-six specific pathogen free( SPF) m ale KM m ice w ere random ly assigned to four groups and each group was further divided into seven subgroups, i. e. three LBP groups receiving low,medium, and high doses of LBH( LBP-L, LBP-M, and LBP-H groups) and three combination groups receiving low, m edium, and high doses of LBP com bined with TP, and a normal saline( N S) group as the control group with 12 m ice in each group. Mice were gavaged with LBP solution or LBP com bined with TP solution at 0. 2m L /10 g body weight daily for 15 d. Then load swimming tests w ere conducted and biochem ical indexes such as hepatic glycogen, serum urea nitrogen, and blood lactic acid were m easured. Results LBP-M, LBP-H, C-L,C-M, and C-H groups had prolonged load swimming time( P 〈0. 05) and LBP-L, LBP-M, LBP-H, C-L,C-M, and C-H groups had elevated hepatic glycogen level( P 〈0. 05) and reduced serum urea nitrogen and blood lactic acid levels( P 〈0. 05) as com pared with the N S group. The com bination groups had prolonged load swimming time, elevated hepatic glycogen level, and reduced serum urea nitrogen and blood lactic acid levels as com pared with the LBP group. The hepatic glycogen level in C-L group w as significantly higher than that in LBP-L group( P = 0. 027) and the blood lactic acid level in C-M group was significantly lower than that in LBPM group( P = 0. 006). Conclusion LBP and LBP com bined with TP can relieve the physical fatigue in micewithout affecting the m ouse body weight. The mechanism maybe associated with increased hepatic glycogen storage, reduced catabolism for proteins, reduced serum urea nitrogen level, increased phosphofructokinase activity, and decreased blood lactic acid level.
