摘要
目的:探讨电针"丰隆"穴对高脂血症模型大鼠血液流变学及炎症相关因子单核细胞趋化蛋白-1(MCP-1)、白细胞介素-1β(IL-1β)、干扰素-γ(IFN-γ)、白细胞介素-10(IL-10)含量的影响,明确电针在治疗高脂血症中的作用。方法:成年雄性SD大鼠40只,每组10只,随机分为空白对照组(A组)、模型对照组(B组)、电针1组(C组)、电针2组(D组),A组给予普通饲料喂养60d,B组给予高脂饲料喂养60d,C组和D组前30d均采用高脂饲料喂养,后30d分别采用高脂饲料喂养+电针治疗、普通饲料喂养+电针治疗;"丰隆"穴取大鼠膝关节外侧后三里下1mm处,用32#1寸针灸针,直刺5mm后接韩式穴位神经刺激仪(30min,2/100 Hz,2mA)。治疗结束后,测定各组大鼠的血脂含量(TC、LDL-C、HDL-C)和血液流变学指标;再分离大鼠腹腔巨噬细胞,检测其中MCP-1、IL-1β、IFN-γ、IL-10的含量。结果:与空白对照组比较,TC、LDL-C明显上升(P<0.01),HDL-C明显下降(P<0.01);与模型对照组比较,大鼠血清TC、LDL-C均有显著下降(P<0.01)。与空白对照组比较,模型对照组血液流变学各项指标均有明显差异性(P<0.05,P<0.01),而电针2组与空白对照组比较无明显的差异性(P>0.05)。与模型对照组比较,电针1组和电针2组血液流变学指标均明显下降,以电针2组更为显著(P<0.01)。与空白对照组比较,模型对照组炎症因子MCP-1、IL-1β、IFN-γ均有显著性升高(P<0.01),而抗炎因子IL-10有显著下降(P<0.01)。与空白对照组比较,电针1组除了MCP-1有显著差异性外(P<0.01),其余各项比较均无明显差异性(P>0.05)。与模型对照组比较,电针1组各炎症因子均有明显减少(P<0.05),而电针2组则减少更为显著(P<0.01)。电针1组与2组的IL-10含量较模型对照组均有显著性升高(P<0.01)。结论:电针丰隆穴能够明显下调高脂血症大鼠血浆中的TC、LDL-C水平及血液流变学指标,减少炎症因子MCP-1、IL-1β、IFN-γ并增加抗炎因子IL-10的含量,从而达到对高脂血症的治疗作用。
Objective: To observe the effect of electroacupuncture (EA) at the Fenglong pointon hemo- rheology and inflammatory factors MCP-1, IL-1β, and IFN-γ of rats with hyperlipemia, and to study the role of electroacupuncture in treating HLP. Methods. Forty healthy male SD rats were selected and randomly divided into four groups, the blank control group (Group A), the hyperlipemia model group (Group B), electroacupuncture group 1 (Group C, high fat diet+EA), and the electroacupuncture group 2 (Group D, normal diet+EA), with 10 rats in each. Blood lipids and blood rheology indicators were assayed. The macrophages were isolated for determination of MCP-1, IL-1β, and IFN-7 by flow cytometry (FCM). Results. Compared with Group A, TC and LDL-C of hyperlipemia rats were much higher (P〈0.01), HDL-C decreased significantly (P〈 0.01). After EA treatment, the contents of TC and LDL-C significantly decreased(P〈0.01). Compared with those in Group A, hemorheology indexes of the Group B had obvious difference (P〈0.05, P〈0.01), while those in Group D had no significant difference (P〉0.05). Compared with Group B, hemorheology had significantly increased in Group C and Group D, especial- ly in the Group D (P〈0.01). Compared with Group A, the MCP-1, IL-1β, and IFN-γ of Group B significantly increased (P〈0.01), while anti-inflammatory factor IL-10 decreased significantly (P〈0.01). Compared with Group B, except MCP-1, the other inflammatory factors showed no obvious difference (P〉0.05). Compared with those of Group A, the inflammatory factors of Group C and D obviously decreased (P〈0.05 and P〈0.01, respectively). Compared with that of Group B, the IL-10 of GC and GD significantly increased (P〈0.01). Conclusion. Electroacupuncture at the Fenglong point can decrease the changes in TC, LDL-C, and hemorheology in rat with hyperlipemia and reduce the content of MCP-1, IL-1β, and IFN-γ, while increasing the content of IL-10, so as to achieve therapeutic effect on hyperlipidemia.
