摘要
改良巨噬细胞的分离与培养方法,为体外研究巨噬细胞提供一种切实可行的实验手段。首先以无血清DMEM培养液灌洗小鼠腹腔约8—10 min,吸出灌洗液于含10%胎牛血清的DMEM培养液中培养。通过细胞形态观察;细胞内酸性磷酸酶及非特异性酯酶表达的测定;细胞吞噬墨汁颗粒功能的检测进行鉴定。最后获得了大量高纯度的细胞,具有巨噬细胞的形态特征,细胞内有巨噬细胞的水解酶类,以及有良好的吞噬功能。实验证明:该方法是一种简易而实用的体外分离培养腹腔巨噬细胞的方法。
Objective： For study on macrophage, to establish a convenient method for separation and cultivation of macrophage. Method： Abdominal cavity of were douched using DMEM medium without serum, and the douche to contain cells was drawn out and the cells were cultured in DMEM medium with 10 % fetal bovine serum. The cells were identified by morphological observation, endocellular enzyme and phagocytosis. Results： The obtained cells were highly purified macrophage with morphologic characteristics of the macrophage in organism, favourable phagocytosis, acid phosphatase （ACP） and a-acetic acid naphthol esterase（α-ANE） activity. Conclusion： This was a simple and pragmatic method for separation and cultivation of rat peritoneal macrophage.
