人类白细胞抗原、信号转导和转录激活因子、整合素与自身免疫病的相关性研究进展 被引量:9

Progress of study on the correlation of human leukocyte antigen, signal transduction and transcription activator and integrins with autoimmune disease

摘要 人类白细胞抗原(HLA)、整合素、信号转导和转录激活因子(STAT)与自身免疫病发生、发展有着密切的联系。自身免疫病是一类原因复杂、临床表现多样、严重危害人类健康的疾病,多为慢性病,任何年龄、性别均可发病,以女性为多见;常累及多器官、多系统,反复发作、慢性迁延。患者血清中常可检出高浓度的自身抗体和/或与自身组织成分起反应的致敏淋巴细胞,并发生免疫效应病理性应答,病理特征常为淋巴细胞和浆细胞浸润等两方面的慢性炎症。基于该类疾病发病机制中不同环节的特异性生物治疗已成为研究热点,并有可能发展为临床治疗的趋势,如针对T细胞和B细胞的生物学治疗,抑制T细胞活化和T-B细胞间相互作用的生物制剂,针对细胞因子的生物制剂,自身免疫病治疗性肽疫苗的研究等。近年来,针对HLA、STAT和整合素的分子功能,国内外的研究结果发现它们三者与多种自身免疫性疾病有高度相关性。对HLA、STAT和整合素与各种自身免疫病的相关性进行归纳分析,为该类疾病的及早诊断和基因防治提供系统性的理论基础和更为开阔的解决方案,具有重大意义。 HLA, Integrins, signal transduction and STAT have a close connection with the onset and development of autoimmune disease. Autoimmune disease is the chronic disease which has complicate causes, various clinical manifestations and serious harm to human health. People of any gender or any age can he infected, especially women; multiple organs and systems damaging often occur, along with repeated attack and chronic persistent. High-concentration au- toantibodies and sensitized lymphocyte which could react with autologous tissue could be detected in patients" serum. They could cause pathological immune response and pathological characteristics often acted as chronic inflammation like lymphocyte and plasmocyte infiltration. Specific biotherapy in different links has been a research focus based on pathogenesis of autoimmune disease and it's possible to be the trend of clinical treatment. For example, biological therapy for T cell and B cell, biological preparation to inhibit T cell activation and T-B cell collaboration, biological preparation for cytokine and research on autoimmune disease therapeutic peptide vaccine. In recent years, HLA, STAT and integrin have been found to have high correlation with many kinds of autoimmune disease according to the research results about their molecular function at home and abroad. The inductive analysis of the correlation of HLA, STAT, inte-grin with all kinds of autoimmune disease, which provide systematic theory foundation and more solutions for early diagnosis and gene therapy, is of great significance.
