摘要
目的:探讨槲皮素(Que)对高糖培养海马神经元的保护作用及其作用机制。方法:原代培养新生大鼠海马神经元3d,随机分为正常对照组、高糖组、阳性对照组及Que 2.5μmol/L(Q2.5)、Que 5μmol/L(Q5)、Que 7.5μmol/L(Q7.5)组,不同条件培养72h后,用流式细胞仪检测神经元凋亡、Western Blot检测GRP78、Caspase-12的表达。结果:与正常对照组比较,高糖组、Que三组及阳性对照组的神经元凋亡率及GRP78、Caspase-12表达均显著增加(P<0.01)。与高糖组比较,阳性对照组和Que各组神经元凋亡率均显著下降(P<0.01,P<0.05);Que各组的GRP78和Caspase-12的表达显著下降(P<0.01),其中Q5和Q7.5组下降尤为显著,且两组比较无统计学差异;与阳性对照组比较,Q5和Q7.5组的神经元凋亡率和抑制Caspase-12表达的作用均无显著性差异。结论:槲皮素能够减轻高糖培养海马神经元凋亡,在此情况下,GRP78、Caspase-12表达均下降,提示槲皮素抑制海马神经元凋亡的作用机制可能与内质网应激有关。
Objective: To explore the protective effect and mechanism of quercetin on hippocampal neurons of Sprague Dawley (SD) rats cultured with high glucose. Methods: Hippocampus were obtained from newborn SD rats, and then separated and purified. Hippocampal neurons were divided into control group, high glucose group, positive control group, Que 2.5μmol/L group (Q2.5), Que 5μmol/L group (Q5) and Que 7.5μmol/L (Q7.5). Hippocampal neurons were cultured in different conditions for 72h. Flow cytometry was used to test the apoptosis of neurons. The expression of GRP78 and Caspase-12 was detected by using western blot. Results: Compared with the control group, the apoptosis of neurons and expression of GRP78 and Caspase-12 of high glucose group, Q2.5, Q5, Q7.5 and positive control group were all increased significantly (P〈0.01). Compared with high glucose group, the apoptosis of neurons of positive control group, Q2.5, Q5 and Q7.5 were decreased significantly (P〈0.01, P〈0.05) and the expression of GRP78 and Caspase-12 of Q2.5, Q5 and Q7.5 was decreased significantly (P〈0.01), especially the Q5 and Q7, and the difference between the two groups was not significant. The difference in apoptosis of neurons and Caspase-12 among positive control group, Q5 and Q7.5 was not significant. Conclusion: Quercetin could reduce apoptosis and expression of GRP78 and Caspase-12 of neurons which cultured in high glucose, which showed that the mechanism of quercetin in hippocampal neurons apoptosis inhibition might be related with endoplasmic reticulum stress.
