摘要
利用基因枪技术 ,将小麦土传花叶病毒外壳蛋白基因CWMV CP1和筛选基因bar导入扬麦 15 8,获得14 5株抗Bialaphos再生植株 ;PCR Southern分析 ,其中 2 1株为阳性植株 ,转化率达到 0 .99% ;T1代植株的PCR Southern、单酶切和双酶切Southern杂交 ,证明外源抗性基因已经完整地整合到小麦基因组中 ;T1代植株分离比CP1+ ∶CP1-为 1∶1.3,偏离孟德尔分离定律 ;T2 代植株总RNA ,RT PCR的试验结果表明CWMV
CWMV CP1 target gene and bar of selection gene were transferred to Yangmai158 by bombardment. Total of 145 plants resistant to 3-5mg/L Bialaphos were obtained. By PCR Southern test in T 0 generation, 21 plants were identified to be positive. The transformation frequency is 0.99%; For tracing the alien gene, the T 1 plants were tested by PCR and Southern blotting. The results demonstrated that the alien resistance gene was integrated into the wheat genome. But the ratio of T 1 plants for CP1 +∶CP1 - is 1∶1.3. They diverged from Mendel′s Law;RT PCR of T 2 plants showed that the alien genes CWMV CP1 had expressed stably.
