摘要
采用农杆菌介导法转化小麦品种G5 4授粉 1 0d后的幼胚 ,经 5‰和 2‰PPT筛选获得 83株正常再生植株。PCR及Southern杂交检测证明其中 8株再生苗为转bar基因植株 ,这些植株对除草剂Basta的抗性明显提高。实验结果还证明 ,高糖浓度的培养基对愈伤组织诱导、植株再生及生根都有显著的促进作用 ;在感染液和共培养基中添加乙酰丁香酮有利于转化株的筛选。
A herbicide-resistant bar gene was introduced into wheat (Triticum aestivum L.) cv.G54 by Agrobacterium infection of immature embryos 10 d after pollination. MB and MS media were used for various steps of culture and transformation with additions of phytohormones, sugar, AS or PPT(Table 1). Eighty-three seedlings were regenerated from 5‰ PPT screening. Eight transgenic plants were identified by both PCR amplification and Southern blot hybridization(Figs.4,5). The results of the study indicates that high concentrations sugars (glucose and sucrose) are effective in inducing callus production, seedling regeneration and rooting of regenerated shoots(Table 3). It has also confirmed that the addition of acetosyringone (AS) in bacteria culture and co-culture media may facilitate the selection of transgenic plants(Table 2).
