摘要
目的探讨大鼠肝癌免疫微环境中T细胞及其亚群的改变;研究胸腺五肽(TP-5)对大鼠肝癌免疫微环境中T细胞及其亚群的影响。方法将60只健康雄性Wistar大鼠随机分成4组:正常对照组:建模和给药周期内一切操作均以等量生理盐水替代药物;模型对照组:建立肝癌模型后,给药期以生理盐水替代TP-5;给药Ⅰ组:建立肝癌模型后,给药期给TP-5,0.25 mg/次,1次/d;给药Ⅱ组:建成肝癌模型后,给药期给TP-5,0.5 mg/次,1次/d。流式细胞术检测肝组织中T细胞及其亚群的改变。结果模型组肝组织中CD3+T细胞、CD4+T细胞、CD4+CD25+调节性T细胞的比例均高于相应的正常对照组(P<0.001),CD8+T细胞在模型组肝组织中的比例低于相应的正常对照组(P<0.001)。TP-5治疗后给药Ⅰ、Ⅱ组的CD3+T细胞、CD4+T细胞、CD8+T细胞、CD4+CD25+Treg细胞与模型对照组间有明显差异(P<0.05),且明显趋于正常(与正常对照组比P>0.05)。结论肝癌免疫微环境中CD3+T细胞、CD4+T细胞以及对免疫起抑制作用的CD4+CD25+Treg细胞比例增高,但在抗肿瘤反应中起主要作用的CD8+T细胞比例却明显下降,导致肝癌免疫微环境处于抑制状态;TP-5对大鼠肝癌免疫微环境有改善作用。
Objective To probe the changes of T-lymphocyte and its subgroups in immune micro-environment of hepatocellular carcinoma in rats; To investigate the effect of Thymopentin in treatment of the immune micro-environment of hepatocellular carcinoma. Methods Sixty wistar rats were randomly divided into four groups: normal control group, hepatocellular carcinoma group, hepatocellular carcinoma with daily peritoneal injection of TP-5 (groupⅠ), hepatocellular carcinoma with ip TP-5 every days ( group Ⅱ) . The two treatment groups received the different consistency of TP-5 through the experiment period. Their Tlymphocyte subgroups in liver tissues were measured by flow cytometry. Results The percentage of CD3+T Cells, CD4+T Cells, CD4+CD25+ regulation T cells in hepatocellular carcinoma group was significantly higher than that in normal control group (P〈0.001, P〈0.001, P〈0.001), on the contrary, the percentage of CD8+T cells in hepatocellular carcinoma group was lower than the corresponding in normal control group (P〈0.001). The percentage of CD3+T Cells, CD4+T Cells, CD8+T Cells in groupⅠor groupⅡwas significantly different from that of hepatocellular carcinoma group (P〈0.05), while the percentage in group Ⅱwas not different from that of normal control group (P〉0.05). Conclusions HCC tissues had higher percentages of CD3+T and CD4+T cells compared with the normal liver tissues, but the proportion of CD8+ T cells which played an important role at the anti-tumor immune response was significantly decreased, while the proportion of CD4+CD25+Treg cells which exerted inhibitory effects on the immune response increased, resulting in the suppressive immune micro-environment in HCC. TP-5 showed a better efficacy in treatment of the immune micro-environment of hepatocellular carcinoma.
