摘要
通过检测塔里木兔Lepus yarcandensis肺脏中水通道蛋白(aquaporin,AQP)1、AQP3及AQP4的表达和分布情况,以探讨水通道蛋白在干旱区动物水液代谢中的作用。采用常规HE染色观察肺组织学结构,采用免疫组织化学检测AQP1、AQP3及AQP4在肺脏中的分布位置及表达。结果表明,AQP1分布在支气管上皮细胞,肺泡间质毛细血管内皮细胞以及肺泡上皮(Ⅰ、Ⅱ型)细胞。AQP3分布在小气管上皮顶质膜和大气道上皮细胞基底膜。AQP4分布在小气管和大气道上皮细胞基底膜。AQP1、AQP3及AQP4在肺中表达的强弱关系为AQP1>AQP4>AQP3。这些结果说明,水通道蛋白在塔里木兔肺泡腔、肺组织间隙及毛细血管腔之间水的转运中很可能起着很重要的作用。同时对吸入空气的湿润和呼出气体中水分的重吸收也具有重要意义。
We detected the expression of aquaporin (AQP) 1, AQP3 and AQP4 in lung of yarkand hares (Lepus yarcandensis) to investigate the role of AQP in animal water metabolism in arid area. The histological structure of lungs was observed by using HE staining, and the expression of AQP1, AQP3 and AQP4 were detected by using the method of immunohistochemistpy. The results showed that AQP1 was localized in bronchioles, blood vessels and pulmonary alveoluses; AQP3 was localized in apical membrane and basal membrane of epithelial cells in bronchioles ; AQP4 was observed at the epithelial cells basal membrane of bronchioles. The expression levels of AQP1, AQP3 and AQP4 in lung were AQP1 〉 AQP4 〉 AQP3. These results suggested that AQP may play a key role in water transport of alveolar cavity, pulmonary interstitial and capillary cavity in yarkand hares. Furthermore, this protein is important to air wetness and reabsoqption of water.
