摘要
目的研究水通道蛋白-2(AQP-2)在小鼠集合管发生发育中的表达规律并探讨其作用。方法采用免疫组织化学和细胞图像分析系统测定灰度值法检测不同时期AQP-2的表达。结果AQP-2在孕16d的胎鼠检测不到,而在孕17d的胎鼠可以明显的检测到,并随着年龄的增长逐渐增加,在生后3周左右达到稳定水平。结论AQP-2在孕17d的小鼠集合管主细胞的顶浆膜和胞浆内出现,其着色强度随年龄的增加而加强,到成鼠时达到稳定水平,推测可能与尿液的浓缩能力有关。
Objective To observe the expression and the rules of AQP - 2 in mouse collecting ducts during development and maturation. Methods The expression of AQP - 2 was examined by immunohistochemistry and analysed by a computerized densitometry. Results AQP - 2 was hardly detectable in 16 - day gestation fetuses. But that was apparently noticed in 17 - day fetuses and was enhanced gradually with age to reach aplateau at 3 week. Conclusions AQP - 2 protein appeared first at the apical side and cytoplasm of collecting duct principle cells in 17 - day fetuses. The staining intensity at the site increased with age, and reach aplateau in mature mouse.The progressive induction of AQP-2 expression in the first 3 week after birth is presumed to contribute to the maturation of urinary concentrating capacity during the kidney development.
