摘要
目的研究虎金丸对肝纤维化大鼠Nrf2/Bach1蛋白表达的影响,初步探讨虎金丸调整机体氧化应激抗肝纤维化的作用机制。方法以CCl4诱导肝纤维化大鼠动物模型,采用Masson胶原染色肝脏纤维化,分光光度法检测SOD、GSH-Px含量,免疫组化法测检Nrf2、Bach1的蛋白量。结果从Masson染色得出,虎金丸能减轻CCL4所致的肝纤维化,显著降低胶原纤维面密度;与模型组比较,虎金丸组SOD、GSH等抗氧化酶的含量显著升高(P<0.01);虎金丸药物的使用显著降低了肝细胞组织匀浆中Nrf2的表达水平、提高了Bach1的表达水平(P<0.01)。结论虎金丸可调控肝细胞组织匀浆中Nrf2/Bach1蛋白的表达,促进机体氧化应激的调整与抗肝纤维化。
Objective To investigate the effects of Hujin pills on the expression of Nrf2/Bachl proteins of rats with liver fibrosis,and to explore the mechanism of Hujin pills in regulating oxidative stress for anti-hepatic fibrosis. Methods The rat models with liver fibrosis were induced by CC14.Masson collagen was used to stain the liver fibrosis.SOD and GSH-Px were measured by spectrophotometry.Nrf2 and Bachl proteins were measured by immunohistochemistry. Results The masson staining experiment shows that Hujin pill could significantly reduce the collagen fibers surface density(P 〈 0.01) therefore alleviate the liver fibrosis induced by CCL4.Compared with model group, the Hujin pill group saw an significant increase in the Content of SOD, GSH and other antioxidant enzymes in the liver homogenate(P 〈 0.01); The administration of Hujin Pills contributed to a decline of Nrf2 expression and an increase of Bachl expression level in liver cell cytoplasm. Conclusion Hujin pills can adjust the level and expression of Nrf2/Bachl proteins in the liver homogenate as well as improve the adjustment of oxidative stress and anti-hepatic fibrosis.
