双虎清肝颗粒对四氯化碳诱发肝纤维化大鼠血清TNF-α、IL-6、IL-10的影响 被引量:2

Effects of Shuanghuqinggan granule on serum levels of TNF-α,IL-6 and IL-10 in hepatic fibrosis induced by carbon tetrachloride in rats

摘要 目的探讨双虎清肝颗粒对四氯化碳诱发肝纤维化大鼠血清TNF-α、IL-6、IL-10的影响。方法清洁级Sprague-Dawley大鼠60只,随机分成6组,A组(正常对照组)、B组(模型组)、C1组(双虎清肝颗粒小剂量组)、C2组(双虎清肝颗粒中剂量组)、C3组(双虎清肝颗粒大剂量组)、D组(水飞蓟素对照组)。除正常对照组外,均经皮下注射400 mL/L四氯化碳8 wk,后各治疗组给予不同剂量双虎清肝颗粒干预8 wk,阳性对照组给予水飞蓟素干预8 wk,股动脉取血,ELISA法检测大鼠血清TNF-α、IL-6、IL-10。结果TNF-α:与B组比较,A组P<0.01,C1组、C2组、C3组、D组P<0.05;IL-6:与B组比较,A组P<0.01,C1组、C2组、C3组、D组P<0.05;IL-10:与B组比较,A组P<0.01,C1组、C2组、C3组、D组P<0.05。结论双虎清肝颗粒能够通过降低大鼠血清TNF-α、IL-6、提高大鼠血清IL-10而发挥抗肝纤维化作用,并且量效关系明显。 Objective To investigate the ei'fects of Shuanghuqinggan granule on serum levels of TNF-α, IL-6 and IL-10 in hepatic fibrosis induced by carbon tetrachloride in rats. Methods 60 Sprague-Dawley rats were divided into 6 groups, including normal control group (A),model group (B),Shuanghu qinggan granule low (C1),medium (C2),high (C3) dose group and silymarin contrast group (D). The rats of B,C1 ,C2,C3 and D were subcutaneously injected by carbon tetrachloride twice a week for 8 weeks. The rats of C1,C2,C3 were administered with Shuanghuqinggan granule at a dose of 5 g/kg body weight (C3) ,2.5 g/kg (C2) and 1.2 g/kg (Cl) for 8 weeks. The rats of D were administered with silymarin at a dose of 50 mg/kg body weight. The changes of serum levels of TNF-α, IL-6 and IL-10 were measured with ELISA. Results Compared with those of group B, the serum levels of TNF-u and IL-6 were significantly decreased in group A, C1, C2, C3 and D (P〈0.05). Compared with those of group B, the serum levels of IL-10 were significantly increased in group A, C1, C2, C3 and D (P〈0.05). Conclusion Shuanghuqinggan granule can reduce hepatic fibrosis induced by carbon tetrachloride in rats and its mechanisms maybe involve in TNF-α, IL-6 and IL-10.
