摘要
目的通过化学合成靶向Survivin的SiRNA转染CNE-2细胞株的方法来探讨以Survivin为靶点的基因干预手段对鼻咽癌细胞生长抑制作用,为鼻咽癌的基因治疗提供新的实验依据。方法通过生物合成Survivin特异性siRNA转染CNE-2细胞,采用Western blotting检测Survivin在干扰后蛋白的表达情况,并筛选出干扰效果最好的siRNA序列转染细胞进行细胞生长抑制试验(CCK-8实验)检测其对细胞生长抑制作用。结果转染siRNA后,CNE-2细胞中survivin表达明显下降,筛选出转染1号序列的实验组CNE-2细胞进行CCK-8实验,结果显示,其细胞增殖明显抑制。结论靶向Survivin的siRNA转染人鼻咽癌CNE-2细胞有效抑制鼻咽癌CNE-2细胞Survivin表达,细胞增殖受到抑制。
Objective To explore the inhibiting effect of siRNA transfection against Survivin on the growth of nasopharyngeal carci-noma cells. Methods The chemically synthesized siRNA against Survivin gene was transfected to CNE-2 cells;the Western blotting was applied in detecting Survivin protein expression in CNE-2 cells, and those CNE-2 cells with better interference effect of siRNA se-quences were filtered out for cell growth inhibition test (CCK-8 experiments) and their inhibiting effect on cell growth was then detect-ed. Results The expression of Survivin in CNE-2 cells after SiRNA transfection was significantly decreased; CCK-8 experiment was made to the filtered out CNE-2 cells of the 1st sequence transfected, which indicated an obvious inhibition on cell proliferation. Conclu-sions Human nasopharyngeal carcinoma CNE-2 cells with transfection of siRNA targeting at Survivincan can effectively inhibit the Sur-vivin expression of CNE-2 cells, and the cell proliferation would be inhibited as well.
