摘要
目的体外观察高压氧对鼻咽癌CNE-2细胞放射敏感性的影响。方法将鼻咽癌CNE-2细胞随机分为A、B、C、D四组。A组:细胞用0.22MPa高压氧处理60min后立即进行2Gy放疗;B组:细胞只应用0.22MPa高压氧处理60min;C组:细胞只进行2Gy放疗处理;D组:细胞不作任何处理。应用MTT法检测各组细胞生长抑制率,PI染色检测各组细胞死亡率,流式细胞计数各组细胞凋亡率。结果 A、B、C3组细胞生长抑制率分别为(18.67±4.54)%,(7.47±4.88)%,(14.06±9.39)%,A组与B组和C组比较差异有统计学意义(P<0.05);A、B、C、D4组死亡率分别为(11.70±1.45)%,(4.00±0.56)%,(8.43±0.83)%,(1.43±0.51)%。A与B、C、D组比较差异有统计学意义(P<0.05);A、B、C、D4组凋亡率分别为4.37±1.12、2.19±0.35、2.70±0.38、1.65±0.20,A与B、C、D组比较差异有统计学意义(P<0.05)。结论高压氧对鼻咽癌CNE-2细胞有较强的放射增敏作用。
Objective To investigate the sensitization effect of hyperbaric oxygenation on radiotherapy of nasopharyngeal carcinoma CNE-2 cells. Methods Nasopharyngeal carcinoma CNE-2 cells were divided randomly into groupA , B , C and D. The cells of group A received hyperbaric oxygenation of 0.22 MPa for 60 min, and were then given radiation of 2 Gy. The ceils of group B received hyperbaric oxygenation as the same as that of group A but without radiotherapy. The cells of group C were radiated 2 Gy only. Those of group D received no treatment and were selected as control. MTT method was used fro detecting the cellular growth inhibiting rates. The cell death rates were observed under PI staining and the cell apoptosis was analyzed with flow cytometry ( FCM ) . Results The cellular growth inhibiting rates of group A, B and C were 18. 67 ± 4.54 % , 7.47 ±4.88% and 14.06 ± 9. 39% respectively, and the difference between group A and group B and C was statisticallly significant ( P 〈0.05). The cell death rates of group A,B,C and D were 11.70 ± 1.45% , 4.00 ± 0. 56% , 8. 43 ± 0. 83% , and 1. 43 ± 0. 51% respectively with a statisticaUly significant difference between group A and B,C,D ( P 〈0.05). And the apoptosis rates were 4.37 ± 1. 12, 2. 19 ±0.35, 2.70 ±0. 38, and 1.65 ±0.20 in group A, B, C and D, and the difference between group A and B. C. D was also statistically significant ( P 〈 0. 05 ). Conculsion Hyperbaric oxygenation could sensitize the radiotherapy effect on nasopharyngeal carcinoma CNE-2 cells.
