摘要
目的:优选大孔吸附树脂纯化薄荷总酚酸的工艺条件。方法:以总酚酸含量和吸附-解吸率为指标,考察5种不同大孔吸附树脂对总酚酸的吸附能力,确定最佳树脂型号,并优化其纯化工艺条件。结果:选用HPD-400型大孔吸附树脂,最佳工艺条件为上样液质量浓度0.1 g.mL-1,上样液pH 3,上样量6 BV,树脂柱径高比1∶9,2 BV 20%乙醇洗脱除杂,流速2BV.h-1,3 BV 70%乙醇以4 BV.h-1流速洗脱,收集洗脱液,薄荷总酚酸纯度可达53%。结论:所建立的工艺分离效果良好,操作简单,重复性好,可用于分离富集薄荷总酚酸。
Objective: To optimize purification technology of total phenolic acid from Mentha macroporous resin. Method: With the content of total phenolic acid and adsorption-resolution rate as indexes,optimum resin model was determined by investigating adsorption capacity of five different macroporous resin on total phenolic acid, and optimized its purification technology conditions. Result: HPD-400 macroporous resin was selected, Optimum purification conditions were as follow: The concentration of sample liquid was 0. 1 g ·mL^-1 and then turn to pH 3 with acetic acid,the amount of sample 6 BV, ratio of diameter-height 1 : 9, 2 BV 20% ethanol to wipe off impurity with flow velocity of 2 BV·h^-1,3 BV 70% ethanol to elute with 4 BV ·h ^-1, eluate collected, purity of total phenolic acid 53%. Conclusion: Optimized purification technology was fit for separating and easy to operate with good reproducibility, it could be used to separate and enrichment total phenolic acid from M. haplocalyx.
