摘要
目的探讨WWOX、p53及Bcl-xl基因在食管鳞癌组织中的表达及其与临床病理学特征的关系。方法应用免疫组织化学SP法检测10例高分化鳞癌、28例中分化鳞癌、14例低分化鳞癌及52例癌旁正常组织中WWOX、p53及Bcl-xl基因的表达并分析其与临床病理指标的相关性。结果 WW0X在食管鳞癌组织中的表达明显降低,与癌旁正常组织相比较其差异有统计学意义(P<0.01),且WWOX的表达与癌组织的分化程度、肿瘤的浸润深度及淋巴结转移情况密切相关(P<0.05),在低分化鳞癌中的表达低于高、中分化鳞癌。p53(突变型)基因在食管癌组织中的表达明显高于癌旁正常组织,二者比较其差异具有统计学意义(P<0.05),其表达与癌组织的分化程度、肿瘤的浸润深度及淋巴结转移情况密切相关(P<0.05),以上两种基因的表达与患者的年龄和性别无关(P>0.05)。Bcl-xl基因在食管鳞癌组织中的表达高于癌旁正常组织(P<0.05),其表达与癌组织的分化程度有关(P<0.05),与肿瘤的浸润深度及淋巴结转移情况及患者的年龄性别无关(P>0.05)。WWOX基因与p53基因及Bcl-xl基因的表达均呈负相关。结论 WWOX在食管鳞癌组织呈低表达,p53(突变型)及Bcl-xl基因在食管鳞癌组织呈高表达。三基因在食管癌的发生、发展及转移过程中有明显的协同作用。
Objective To investigate the expression and clinical significance of WWOX,p53 and Bcl-xl protein in esophageal carcinoma.Methods Immunohistochemical staining methods were used to respectively detect the espression of WWOX,p53 and Bcl-xl protein in 10 cases of high differentiated squamous cell carcinoma,28 cases of medium differentiated squamous cell carcinoma,14 cases of poorly differentiated squamous cell carcinoma and 52 cases of normal esophagus tissue.And the results were analyzed together with clinical pathologic characteristics.Results WWOX in esophageal carcinoma demonstrated obvious low expression,there was significant statistical difference between carcinoma tissues and normal tissues(P 0.01).At the same time,the expression of WWOX demonstrated close relationship with tumor degree of differentiation,tumor invasion depth and lymph node metastasis(P 0.05).Its expression in poorly differentiated squamous cell carcinoma was lower than high differentiated squamous cell carcinoma and medium differentiated squamous cell carcinoma.The expression of p53(mutant type) in esophageal carcinoma was significantly higher than that in normal tissues,there was significant statistical difference between the two groups(P 0.05),and its expression was closely related to the degree of differentiation,tumor invasion depth and lymph node metastasis(P 0.05).The two types of gene expression and the patient's age and gender irrelevant(P 0.05).The expression of Bcl-xl in esophageal carcinoma was significantly higher than that in normal tissues(P 0.05),its expression and the differentiation of the cancer tissue levels(P 0.05),the expression of the above three genes had no obvious relation with the patient's age and gender(P 0.05).WWOX in esophageal carcinoma had negative relation with p53 and Bcl-xl.Conclusion The expression of WWOX in esophageal carcinoma is lower than that in normal tissues,and the expressions of p53 and Bcl-xl in esophageal carcinoma are higher than that in normal tissues.The three genes have obvious synergistic action in generating,developing and transferring process of esophageal carcinoma.
