摘要
目的:探讨WW结构域氧化还原酶基因(WW domain-containing oxidoreductase gene,WWOX)与P73蛋白在皮肤恶性黑色素瘤(cutaneous malignant melanoma,CMM)中的表达及其意义。方法:采用免疫组化法(SP)检测66例CMM、30例交界痣和20例正常皮肤组织蜡块中的WWOX、P73蛋白表达水平。使用SPSSl7.0统计软件包,采用卡方检验和Spearman等级相关分析方法进行统计学分析,检验标准a=0.05,以P<0.05为有统计学意义,χ2分割检验标准0.0167。结果:WWOX在正常皮肤的表达水平明显高于交界痣,其在交界痣的表达水平明显高于在CMM中的表达(P<0.0167);CMM中WWOX的表达水平随肿瘤浸润深度的增加而降低,伴有淋巴结转移者中的表达水平低于不伴有淋巴结转移者,并且差异均具有统计学差异(P<0.05)。P73蛋白在正常皮肤的表达水平低于交界痣,其在交界痣的表达水平低于CMM(P<0.0167);CMM中的P73蛋白表达水平随肿瘤浸润深度的增加而增高,伴有淋巴结转移者中的表达水平高于不伴有淋巴结转移者中的表达水平(P<0.05)。WWOX与P73蛋白在CMM中的表达呈负相关(r=-0.298,P=0.015)。结论:WWOX与P73共同参与调控CMM的发生发展,对其进行联合检测,有助于CMM的早期诊断,为临床治疗提供参考指标,同时也为CMM的基因治疗提供一个新思路。
Objective To study the expression and correlation of WWOX and P73 protein in cutaneous malignant melanoma. Methods Retrospective method was used in the study.The expression level of WWOX and p73 was determined using immunohischemistry staining in 66 cases of cutaneous malignant melanoma,30 cases of junctional nevus and 20 cases of normal skin tissues.The results were analyzed using chi- squared test and Spearman rank correlation test.P〈0.05 is considered statistically significant. P=0.0167 used for chi- squared test.All statistical analyses were performed using SPSS17.0. Results The positive rate of WWOX protein in CMM is significantly lower than that in junction nevus,and the positive rate in junction nevus is significantly lower than that in normal skin(P〈0.0167),with the depth of tumor infiltration increasing,the expression of WWOX in CMM has decreased(P〈0.05),the expression of WWOX in the tissue with lymph node metastasis is significantly lower than that without lymph node metastasis(P〈0.05).The positive rate in normal skin was less lower than that in junction nevus,and the positive rate in junction nevus was less lower than that in CMM(P〈0.0167),with the depth of tumor infiltration increasing,the expression of P73 in CMM has increased(P〈0.05),the expression level of P73 in the tissue with lymph node metastasis is significantly higher than that without lymph node metastasis(P〈0.05). Conclusion WWOX and P73 might play an important role in occurrence and development of CMM,The combined detection of WWOX and P73 might be helpful for early diagnosis of CMM,to provide reference for clinical treatment and supply a new way for gene therapy of CMM.
