摘要
目的:研究不同浓度薏苡仁酯对胃癌BGC-823细胞株的侵袭、迁移能力及CD44、CD133表达的影响。方法:以不同浓度薏苡仁酯作用于BGC-823细胞,MTT法检测细胞增殖抑制率,黏附试验、Transwell小室侵袭迁移实验检测细胞黏附、侵袭、迁移能力,RT-PCR法检测细胞CD44、CD133 mRNA表达水平。结果:薏苡仁酯可抑制BGC-823细胞增殖,其抑制效应呈剂量-时间依赖性;经薏苡仁酯作用后BGC-823细胞的侵袭、迁移及黏附能力降低;CD44、CD133 mRNA的表达与薏苡仁酯浓度亦呈负相关。结论:薏苡仁酯能降低胃癌BGC-823细胞株的侵袭、迁移、黏附能力,其机制可能与通过下调CD44、CD133表达有关。
Objective:To investigate the effect of coixenolide on adhesion,invasion and metastasis of gastric carcinoma BGC-823 cells in vitro and its mechanism.Methods:MTT assay was used to examine the effect of coixenolide on proliferation of BGC-823 cells.Matrigel adhesion assay and Transwell chamber were performed to determine the adhesion,invasion and migratory capacity of the cells.RT-PCR was applied to detect the expression of CD44 and CD133 mRNA in BGC-823 cells.Results:Coixenolide could inhibit the proliferation of BGC-823 cells with obvious dose-dependent and time-dependent effects.Coixenolide significantly inhibited adhesion,invasion and migratory capacity of BGC-823 cells in vitro.After coixenolide treatment for 24 h,the expression level of CD44 and CD133 mRNA was decreased in a dose-dependent way.Conclusions:Coixenolide can inhibit the migration,invasion and adhesion capacity of BGC-823 cells in vitro.The inhibition effect may be related to down-regulating the expression of CD44 and CD133.
