摘要
目的:从基因及蛋白水平探讨中药槐耳调节T辅助淋巴细胞(Th)平衡的机制,为槐耳在临床上治疗缓解期哮喘提供理论依据。方法:将哮喘缓解期儿童外周血单个核细胞(PBMC)分为3组,即空白组、中剂量组和大剂量组,分别与不同浓度的槐耳菌质配液(6g/L)及培养液作用48h;然后测定细胞培养沉淀中白细胞介素-4(IL-4)及干扰素-γ(IFN-γ)mRNA的表达和细胞培养上清中IL-4及IFN-γ的量。结果:在中剂量组和大剂量组中,IL-4mRNA表达明显低于空白组(P<0.05);IFN-γ表达明显高于空白组(P<0.05);IFN-γ/IL-4比值的基因及蛋白水平表达均高于空白组;然而,对于以上4个指标的检测,大剂量组与中剂量组之间无明显差异(P>0.05)。结论:槐耳菌质能够明显增加IFN-γ及减少IL-4mRNA的表达,可调节哮喘儿童缓解期IFN-γ/IL-4比值的失衡,平衡Thl/Th2比值的漂移。
Objective: To explore the mechanism of Trametes Robinioplila on regulating the imbalance of Th1 and Th2 ratio in gene and protein level,and to find the theoretical basis of the fungus for treating asthma in remission stage. Methods: The PBMC from asthmatic children were divided into three groups(blank group, middle-dose group,large-dose group), and then treated respectively with Trametes Robinioplila (0g/L,3g/L,6g/L) and culture medium for 48 hours respectively.The expression of IL-4 and IFN-γ mRNA in cellular precipitation were measured, and the expression of IL-4 and IFN- γ in supernatant were measured too. Results: After PBMC cultured with Trametes Robinioplila for 48 hours, the expressions of IL-4 mRNA, IFN-γ, IFN- γ/IL-4 mRNA ratios and IFN- γ/IL-4 ratios were significantly different(P〈0.05) between drug intervention group and blank group, but there were no markedly differences (P〉0.05) between large-dose group and middle-dose group.Conclusion: Trametes Robinioplila markedly increased the expression of IFN- γ and decreased the expression of IL-4 mRNA, then regulated the imbalance of IFN- γ/IL-4 and Th1/Th2 ratios in asthmatic children in remission stage.
