摘要 目的:建立土荆芥中槲皮素和山柰素的含量测定方法。方法:用20%盐酸水解土荆芥中黄酮类成分得槲皮素和山柰素,并采用 HPLC 法测定槲皮素和山柰素的含量。色谱柱:Hypersil C_(18)(250 mm×4.6 mm,5 μm);流动相:乙腈-0.1%甲酸溶液(40:60);流速:1 mL·min^(-1);检测波长:360 nm。结果:槲皮素和山柰素的线性范围分别为0.0203~0.203μg(r=0.9999)及0.0944~0.944μg(r=0.9998);平均回收率(n=9)分别为99.3%及99.7%。结论:本法简单,重复性好,可用于土荆芥中槲皮素和山柰素含量的测定。 Objective:To develop an HPLC method for determination of quereetin and kaempferol in Herba Chenopodii Ambrosioidis. Methods:The flavones in Herba Chenopodii Ambrosioidis were hydrolyted into quercetin and kaempferol with 20% hydrochloric acid and quercetin and kaempferol were chromatographed on a Hypersil C18 colnmn (250 mm× 4.6 mm,5 μm)and detected at 360 nm. The mobile phase was acetonitrile -0. 1% formic acid solution(40:60) and flow rate was 1 mL · min^-1. Results:The linear range of quereetin and kaempferol were 0. 0203 - 0. 203 μg ( r = 0. 9999 ) ,0. 0944 - 0. 944 μg ( r = 0. 9998 ) respectively. The average recoveries ( n = 9 ) of quercetin and kaempferol were 99.3% ,99.7% respectively. Conclusion:The method is convenient, reliable and repeatable, and can be used for the determination of quercetin and kaempferol in Herba Chenopodii Ambrosioidis.
