摘要
从病鸭翅静脉无菌采集抗凝血,直接镜检和姬姆萨染色镜检,微生物感染呈阳性.从血液中分离出该微生物,用细菌16S rDNA通用引物扩增出约1500bp的条带,经DNA测序得到一条长1499bp的16S rDNA序列,与GeneBank已提交的序列进行BLAST相似性搜索表明该微生物应归属于假单胞菌属(Pseudomonas),并用DNAStar进行同源性分析并构建系统进化树,显示与恶臭假单胞菌(Pseudomonas putida)同源性最近,首次发现恶臭假单胞菌可能感染家鸭.
Blood samples were collected anticoagulatively through wing-vein of sick ducks. Observing blood and Giemsa stained blood by microscope, the results were positive. The genomic DNA of the pathogen isolated was extracted from blood samples. Several 16S rDNA fragments of about 1 500 bp were amplified with bacteria 16S rDNA universal primers, cloned into pMD18-T vector (TaKaRa), and then transformed in the competent cells of E. coli DH5α, several 16S rDNA sequences of 1499 bp were obtained by DNA sequencing. The result of BLAST in GenBank indicated that this pathogen belongs to Pseudomonas. The phylogenic tree basing on 16S rDNA generated by software DNAStar showed that it was most close to Pseudomonas putida. These findings verified the possibility of duck disease may caused by Pseudomonas putida for the first time.
