摘要
为获得油菜抗菌核病的抗源材料,进行了油菜抗菌核病突变体筛选方法的研究.用油菜种子未成熟胚或成熟胚在MS-1(MS+2,4-D2mg/L+IAA0.2mg/L+KT0.1mg/L)培养基上诱导油菜愈伤组织,继代4~5次后用含草酸600mg/L的无Ca2+MS-1培养基处理2~4mm大小的愈伤组织96h,筛选获得的耐毒素愈伤组织移至MS-2(MS+BA1mg/L+IAA0.2mg/L)培养基上获得再生小植株.
The procedure of screening for rapeseed mutants with resistance to stem rot (Sclerotinia sclerotiorum (Lib) De Bary) was proposed.Rapeseed calli were induced from immature or mature embryos on the solid MS medium supplied with 2,4D 2 mg/L,IAA 0.2 mg/L and KT 0.1 mg/L. After subcultured for 4 or 5 generations on the same medium as above the calli were cut into segments in size about 2~4 mm and treated with Ca2+free oxalic acid at the concentration of 600 mg/L for 96 h. The selected resistant calli were transplanted onto the solid MS media supplied with BA 1 mg/L and IAA 0.2 mg/L to regenerate plantlets. Some resistant rapeseed variants were obtained.
