摘要
目的以磁源空间磁场定量计算为依据,探讨不同磁感强度永磁磁场对人脐静脉内皮细胞(ECV304)体外培养的影响及其量效关系。方法选取极面中心磁感强度为93.3,175.3,263.6,320.3和375.4 mT的5组圆片磁源,采用有限元数值法计算其空间磁场,得出磁感强度并以磁感强度等值线表示;将5种强度的磁源分别作用于人脐静脉内皮细胞进行体外培养,用MTT法检测细胞活力(OD值)。结果各加磁组人脐静脉内皮细胞OD值(1.285±0.164,1.251±0.122,1.142±0.176,1.021±0.127,1.015±0.240)均比非加磁对照组OD值(1.411±0.079)下降,且差异均有统计学意义(P<0.05或0.01);总体呈现出磁感强度越强,OD值下降越明显的趋势。结论实验所使用各磁感强度组磁场均对人脐静脉内皮细胞体外增殖有抑制作用,提示磁场可能对增生性瘢痕血管形成有预防和治疗作用。
Objective To study the effect of permanent magnetic fields on human umbilical vein endothelial cells (HUVECs) euhured in vitro, and to explore the dose-effect relationship based on the calculated intensities of space magnetic fields. Methods Five kinds of circular magnets, of which magnetic induction intensity at the polar center were 93.3 mT, 175.3 mT, 263.6 mT, 320.3 mT, 375.4 mT, were chosen. The finite element method was used to calculate the magnetic induction intensity of the magnetic space fields of those magnets. The magnetic induetion intensities were expressed as magnetic induction isolines. Then, the magnets were used to act on HUVECs cultured in vitro, respectively. HUVECs vitality was measured with MTT method. Results The OD values of the five magnetic groups were 1.285 ±0.. 164, 1.251±0.. 122, 1. 142±0.176, 1.021±0.127, 1.015 ± 0.240, respectively and were all significantly lower than that of the control group( 1.411 ± 0.079) ( P 〈 0.05, P 〈 0.01 ). The stronger the magnetic induction, the lower the OD values. Conclusion Magnetic fields have inhibition effect on HUVECs cultured in vitro. This shows that magnetic fields might be able to prevent hypertrophic sear angiogenesis.
