摘要
以苏丹草185的成熟种子为外植体,就不同培养基和激素配比对愈伤组织的诱导率进行研究,筛选出了适合苏丹草185愈伤诱导的培养基。研究中发现,对于苏丹草185种子在相同激素配比条件下,采用MS基本培养基的诱导率高于N6培养基。在相同的基本培养基培养条件下,采用不同激素配比,8个处理组合的诱导率有较大差别。在MS培养基中,2,4-D浓度为2.0mg/L、KT浓度为0.1mg/L时的诱导效果最好;而在N6培养基中,2,4-D浓度为4.0mg/L、KT浓度为0.5mg/L时的诱导效果最好,较易形成胚性愈伤。
Sudan grass 185 was used to study the factors affecting callus induction ratios from different media and factors in sorghum mature seed culture. The results show that the callus induction rate was higher in MS medium than that in N6 medium. There were considerable differences in induction rate and callus quality among 8 treatments. In MS medium, when the concentration of 2,4-D was 2.0 mg/L and the concentration of KT was 0.1 mg/L the callus induction rate was higher and it was easier to form embryogenic callus. By contrast, the callus induction rate in N6 medium was the highest when the concentration of 2,4-D and KT were 4.0 mg/L and 0.5 mg/L respectively.
