摘要
选择计量罐式连续流加补料系统,采用新选育的黑曲霉(Aspergillus niger)Wml-016菌株在50 L自动发酵罐上研究间歇补料对发酵的影响。利用中心复合设计和响应面分析方法,实验得出木薯原料柠檬酸补料发酵的优化条件:发酵到第36 h开始进行4次流加补料,间隔时间为1.5 h,补入DE值为80的木薯水解液和质量浓度为0.04%的(NH4)2SO4。采用上述工艺发酵产酸率达到18.5%,残糖为0.2%,周期仅为64 h,结果明显优于分批发酵。
The effect of fed batch system with the measuring tank on the fermentation of a newly selected strain, Aspergillus niger wml- 016, was studied in a 50 L automatic fermentor. Optimal conditions were obtained from the experiments with methods of central composite design and response surface analysis. The feeding is commenced at 36th hour at intervals of 1.5hours for four times, and the DE of the feeding cassava hydrolyte is 80, while the concentration of feeding (NH4)2SO4 is 0.04 % (w/v). By the above mentioned techmiques, the citric acid productivity is up to 18.5 % ; the residue glucose is 0.2 %, while the fermentation period is only 64 hours. And the process is obviously much better than traditional fermentation.
