摘要
从东亚钳蝎粗毒中分离纯化出蝎毒素Ⅳ,经套管注入侧脑室,用辐射热测痛和屈肌反射检测两种方法,观察蝎毒素Ⅳ的中枢镇痛作用,结果蝎毒索Ⅳ可以明显延长大鼠缩腿潜伏期,并可显著抑制C纤维诱发的屈肌反射。提示蝎毒素Ⅳ具有显著的中枢镇痛作用。
Purified venom component Ⅳ of scorpion (SVC-Ⅳ) were obtained from the crude venom of scorpion Buthus Martensii Karch with CM-Sephadex C-50 and Sephadex G-50 gel filtration. 18 rats were divided into four groups: 3 ug SVC-Ⅳ group (n=5), 6 μg SVC-Ⅳ group (n=5), normal saline control group (n=4), morphia control group (n=4). After 3 ug/10 μl,6μg /10 μl,10 μl saline and 20 μ/10 μl morphia were injected intrathecally into the lateral ventricles of the rats. The analgesic effect of SVC-Ⅳ was investigated with thermal radiation withdrawal reflex and flexor reflex evoked by electrically-stimulating the plantar hind paw.The results are as follow: ①Significant increase of withdrawal reflex latency was observed after the injection of SVC-Ⅳ of two kinds of doses. Compared with the basic withdrawal reflex, the 3 μg SVC-Ⅳ group increased by 33.1%~78.5%,6 μg SVC-Ⅳ group by 31.2%~150% and the normal saline group has little increase. ③In the two SVC-Ⅳ groups, the flexion reflex evoked by C-afferent fibers was markedly depressed, whereas the flexon reflex evoked by A-afferent fibers remained unchanged. In the 3 μg SVC-Ⅳ group, the depression appeared in five minutes and reached the highest in 60 minutes and the rate of depression was 75 per cent; in the 6 μSVC-Ⅳ group, the depression appeared in two minutes and reached the highest in 80 minutes; in the morphia group, the depression appeared intensely and reached the highest in 45 minutes and the rate of depression was 77 per cent, nearly the same with the 3 μg SVC-Ⅳ group.The results indicated that SVC-Ⅳ had strong analges effect when administrated to the central nervous system. To a certain extent, the analgesic effect was dose-related.
