地塞米松诱导促进BMP-2基因转染的兔MSCs的成骨转化 被引量:1

摘要 目的: 研究地塞米松诱导对BMP 2基因修饰的rMSCs的增殖和分化表型的影响.方法: 将地塞米松诱导和未经诱导的兔MSCs分别转导BMP 2基因后,以RT PCR和免疫组化方法检测转导后细胞BMP 2的表达;通过观察细胞生长及碱性磷酸酶活性和骨钙素含量的测定,分析地塞米松诱导对BMP 2基因修饰的rMSCs的增殖和分化表型的影响.结果: 转导后BMP 2基因在两组rMSCs中均有表达,定量分析显示诱导组细胞碱性磷酸酶活性 ( 134 36±8 84 )nkat/L较对照组(104 02±7 83)nkat/L明显增高;诱导组细胞骨钙素含量(14 68±0 73)μg/L较对照组(6 52±1 21)μg/L明显增高.结论: 转导前地塞米松诱导能够促进BMP 2基因修饰的rMSCs的增殖和成骨转化. AIM: To determine the effect of the induction of dexamethasone on the proliferation and differentiation phenotype of BMP-2 modified rabbit mesenchymal stem cells (rMSCs).METHODS: The rMSCs,treated and untreated with dexamethasone,were transfected with BMP-2 gene.The expression of BMP-2 was determined by RT-PCR and immunocytochemical staining.The changes of the proliferation and differentiation phenotype in the cells were observed by cell growth state,alkaline phosphatase (ALP) activity and osteocalcin (OCN) content.RESULTS: The expression of hBMP-2 was confirmed in levels of mRNA and protein.Cell cultures of rMSCs pretreated with dexamethasone showed a higher ALP activity [(134.36±8.84) nkat/L in transfected group versus (104.02± 7.83) nkat/L in control group] and higher osteocalcin content [(14.68±0.73) μg/L in transfected group versus (6.52± 1.21) μg/L in control group].CONCLUSION: Early osteoblastic differentiation induced by dexamethasone can significantly promote BMP-2 modified RMSCs osteoblast conversion.
