摘要
:目的 观察肝素钠对成人骨髓基质细胞增殖及成骨转化的影响。方法 从成人来源的骨髓基质细胞在含不同浓度肝素钠的 16 40培养液中培养 48、72小时 ,MTT比色法观察肝素钠对其增殖的影响 ;采用碱性磷酸酶细胞化学染色 ,计数成骨细胞转化率。结果 肝素钠在 6 0、15 0U/ml时 ,光吸收值明显高于对照组 (P <0 0 1) ,其余浓度组无明显差异 ;对成骨转化率无影响。结论 肝素钠在一定浓度范围内对骨髓基质细胞增殖及成骨转化无抑制作用。
Objective: To observe the effects of heparin on the proliferation and osteogenic transformation of human bone marrow stromal cell(BMSC) Methods: BMSC from adults were cultured for 48 and 72 hours in 1640 media with different concentrations of heparin. MTT method was used to observe the effect of heparin on the cell proliferation The osteogenic transformation rate was calculated by alkaline phosphatase(AKP) cytochemistry stains Results: The potical density values of 60,150 U/ml were significantly higher than those of the control group(P<0 01), the others showed no difference Heparin had no effect on osteogenic transformation of BMSC Conclusion: Heparin in certein concentrations doesnot inhibit the proliferation and osteogenic transformation of BMSC
