VEGF基因转染骨髓间充质干细胞移植治疗对心肌梗死大鼠的影响 被引量:5

摘要 目的探讨血管内皮生长因子(VEGF)基因转染骨髓间充质干细胞(MSCs)移植治疗对大鼠心肌梗死组织修复重建、血管再生及心功能的影响。方法体外分离、培养、纯化大鼠MSCs,将pEGFP-N1/VEGF重组质粒转染MSCs。30只心肌梗死大鼠造模成功4周后随机分为3组,A组(n=10):pEGFP-N1/VEGF重组质粒转染MSCs后行梗死心肌内注射;B组(n=10):大鼠MSCs行梗死心肌内注射;C组(n=10):无血清IMDM培养液行梗死心肌内注射。移植4周后观察移植细胞的分化和新生血管的形成,并通过超声多普勒检测心功能变化。结果 A、B两组大鼠梗死心肌处可见BrdU标记的移植细胞,心肌钙蛋白T染色阳性。A、B两组大鼠MSCs移植后左室射血分数(LVEF)和左室舒张末期内径(LVDd)等指标较移植前均有明显改善(P<0.05),且A组大鼠LVEF和LVDd等指标改善程度要明显好于B组(P<0.05);A、B两组毛细血管数目较C组明显增多(P<0.05),且A组增加幅度更为明显(P<0.05)。结论 VEGF基因转MSCs移植治疗心肌梗死大鼠可通过促进心肌再生和新生血管的形成来修复重建缺血心肌,最终改善心功能。
